To prepare Chuangui huxin patch, a transdermal delivery form containing extraction Ligusticum chuanxiong Hort., Agelica sinensis(Oliv.) Diel, optimum extraction processes for active ingredient of ferulic acid (FA) from these two Chinese herb had been determined by orthogonal experiment. In order to probe the transdermal characters of the active ingredient and the effection of the prescription of Chinese herb on them, the permeability and the influencing factors of FA in the collection were researched by means of drug permeation tests, which performed in K-C diffusion cell through mouse skin in vitro. The effection of permeation enhancers for preparing patch were investigated to find out a better method to enhance the trandermal absorption amount by using drug permeation tests. On the basis of the research, Chuangui huxin patch was prepared.The results showed that the optimum extraction processes of FA of Ligusticum chuanxiong Hort. alcohol extract was: adding 8 times amount of 70% alcohol, refluxing and extracting for 2 times, 1.5 hours each time; the optimum extraction of FA of Agelica sinensis(Oliv.) Diel water extract was: the quantity of water used for extract was 10, 6, 4 times of the quantity of the herb respectively and the extract time was 90min, 60min, 30min respectively. Extract concintration was 1.5:1 and the first concentration of ethanol was 60%.The effective ingredients of FA could penetrate isolated animal skin. There was differences in transdermal absorption of the ingredients between single and herb prescription. The transdermal amount of drugs has been influenced by the permeation enhancer. The permeability of FA has been greatly increased by the permeation enhancer of 2 % water-solubility Azone and 2 % Broneolum.The comprehend evaluation was made on the basis of the characteristics of the patch,then the most perfect formula of the patch was determined finally . The amount of FA had permeated in vitro mouth skin was determined. With the cumulative permeation quantity of FA as the index, 2 % azone is more suitable to act as a permeation enhancer of Chuangui huxin patch.Cutaneous stimulation experiment indicated that it has non-toxicity or non-stimulus to rabbit skin and non-sensitization to Cavia procellus skin.
