Objective: Chemotherapy play an important role in synthetic therapy of breast cancer.Because of multidrug resistance, the effect of chemotherapy is impacted, even the chemotherapy is fail, which could impact the prognosis of patients. For this reason, searching reversal agent for multidrug resistance to enhance the curative effect of chemotherapy is problem to be emergent settled in clinic. At present there are a lot of investigations concerned with retroconversion of multidrug resistance . Now the active agent which can reverse multidrug-resistance are as follow: calcium channel blocker, immunomodulator, calmodulin inhibitor and so on. Because the curative effect is not good, target is single and toxic action is obvious, the reversal agent is infrequently applied in clinic. Chinese traditional drug possesses the good quality of high performance, multitarget and little toxic action, so it has the value of being investigated. To search safe and active reversal agent for multidrug resistance from Chinese traditional drug is a hot spot now. Panaxnotoginseng saponins which has the activity of calcium channel blocker is mainly researched as decoagulant to cure cardio-cerebral vessel disease . This experiment is mainly to study and research that panaxnotoginseng saponins can reverse MCF-7/ADM cell multidrug resistance whether or not.Methods : To measure direct cell toxicant effect of PNS to MCF-7/S(sensitive strain) and MCF-7/ADM (multidrug-resistant strain) and measure toxicant effect of ADM to both strains for resistant index by MTT;to observe the reverse effect of PNS which has screened for nontoxicant concentration to MCF-7/ADM by MTT;to measure expressional positive rate of MDR-1 and P-GP of the both cell after applied PNS by PCR andWESTERN .Results: (1)There is no obvious cell toxicity of PNS to MCF-7/S and MCF-7/ADM cell, when the concentration of PNS is 200ug/ml below. (2)Inhibitive concentration 50 of ADM to MCF-7/S and MCF-7/ADM are(0.62±0.03)ug/ml and(25.03±5.37)ug/ml respectively. Drug-Resistant index of MCF-7/ADM is 40.37.(3)PNS can increased cell toxic effect of ADM, after applied PNS of 50ug/ml,100 ug/ml, 200ug/ml concentration respectively to MCF-7/ADM for 48 hours, inhibitive concentration 50 respectively decline from (25.03±5.37)ug/ml to(17.87±4.27)ug/ml,(13.90±4.63) ug/ml,(11.91±4.69)ug/ml(P<0.01); (4)PNS(50,100,200ug/ ml) can reduced the expression of MDR-1 and P-GP more obviously with the augmentation of dosage.in contrast find no significant effect with verapmil.Conclusions:PNS can partly reverse multidrug-resistance of MCF-7/ADM ,by reducing MCF-7/ADM's expression of MDR-1 and P-GP , which is different from Ver.
