| Objective: Determining the expression of PTEN, MMP-2 and MMP-9 in gastrointestinal stromal tumors (GIST) and analyzing some aspects as following: 1.The relationship between the expression of PTEN, MMP-2 and MMP-9 in GIST and the clinical pathology characteristics of GIST. 2.Analyzing the relationship among PTEN, MMP-2 and MMP-9 and between the biological behaviors of GIST. 3.For the purpose on providing scientific basis for the diagnosis, therapy and prognosis evaluation of the GIST. 4.In order to provide theory basis of effectively adjunctive therapy .Approaching more scientific grouping methods of GIST.Methods: 54 GIST patients whose pathological diagnosis was defined were selected from general surgery department of Shandong provincial hospital from January of the year 2001 to January of the year 2005. We took slices from GIST paraffin blocks preserved in pathological department and determinate the protein expression of PTEN, MMP-2 and MMP-9 in it through immunohistochemical method(S-P method). Experimental data were analyzed together with the clinical pathological datum.Results: 1. The positive expression rates were 72.2%(39/54),85.2%(46/54) and 83.3%(45/54),which were connected with growth pattern, tumor diameter, infiltration and metabasis, nuclear division rating and necrosis of GIST (P<0.05 or P<0.01) , but had nothing to do with patients'age, sex, initial occurring position of the tumor.2.The expression of PTEN and MMP-9 is connected to each other (r=0.63,P<0.01) ; There are relationship between the expression of MMP-2 and MMP-9 (r=0.67,P<0.05) ; There is nothing to do with the expression of PTEN and MMP-2 (r=0.21 ,P>0.05 ) .3. The expression of MMP-2,MMP-9 increased significantly according to the malignant increasing (P<0.01) and associated with pattern of tumor growing,tumor size,and centre necrosis (P<0.01) .4. In univariate analysis, PTEN,MMP-2,MMP-9 were significantly associated with tumor free survival . In multivariate analysis,co-expression of MMP-2 and MMP-9 was unfavorablely related with tumor free survival(RR=2. 487: P<0. 05).Conclusion: 1.There are relationship between PTEN,MMP-2,MMP-9 and the occurrence,development of GIST. PTEN,MMP-2,MMP-9 probably participate in the proliferation. infiltration and metabasis; 2.Probably, PTEN,MMP-2,MMP-9 can influence the infiltration and metabasis of GIST. 3.The detection of PTEN,MMP-2,MMP-9 in GIST is available to the malignant level evaluation, therapy guidance and prognosis evaluation; 4.This study may probably provide adjunctive therapy method and new theory basis of treatment, such as the inhibitation of angiogenesis; 5.The grouping of low and high potential malignancy is more corresponding the biological characteristics of GIST. |
