Background: Aromatic fatty acids, of which phenylacetate(PA) is the prototype, constitute a class of low toxicity drugs with demonstrated antitumor activity in experimental models and in humans.Phenylacetate compounds can induce growth inhibition and differentiation of cancer cells and apoptosis. But single medicine usually can't cure tumor radically, the tumor relapses easily after the medicine-withdrawing, therefore scholars all claim to association with chemotherapy and radiotherapy to raise curative effect. Researches on phenylacetic acid association with chemotherapy functioning on the larynx cancer cell stub have been still reported in our country. Observing the function of phenylacetic acid unites 5 Fu and CDDP to the larynx cancer cell to repress cells-propagation, and induce cell to divide and promote apoptosis compared with single chemotherapy medicine. The Bcl-2 family, consisting of both proapoptotic proteins bax and antiapoptotic proteins bcl-2, is a central regulator of apoptosis acting mainly on the mitochondrial pathway of cell apoptosis.Bax is mvolve in the development and prognosis of cancer. The Bax determines the susceptibility of human cancer cells to chemotherapy, so it is amony proapoptotic proteins to most extensively studied.Objective: Observe the function of Phenylacetic acid united with 5-Fu and CDDP to the larynx cancer cell to promote apoptosis And Bax expression compared with single chemotherapeutic medicine.study the mechanism that the phenylacetic acid enhances the chemotherapy medicine to the larynx cancercell stub apoptosis.Methods: experiments applied to culture in vitro of Hep-2 cells, After on the 24h of culture , Hep-2 were randomly divided into 6 groups: control group ,PA,5-Fu,CDDP,PA with 5-Fu,PA with CDDP. Apply to acridine orange staining , immunohistochemistry, observe the function of Phenylacetic acid united with 5 Fu and CDDP to the larynx cancer cell to promote apoptosis and expression of bax comparing with single chemotherapy medicine. study the mechanism that the Phenylacetic acid enhances the chemotherapy medicine to the larynx cancer cell stub apoptosis.Results:1. acridine orange staining:Control group:The apoptosis rate was (0.79±0.09)%.Under fluorescent microscope it was observed that mostly adqulis tumor cell of virent fluorescence,and infrequently yellowish green cell of apoptosis. 5-Fu group:The apoptosis rate was (3.47±0.87)%. Onder fluorescence microscope it was observed that considerable tumor cells of virent fluorescence,and scattering lesser yellowish green cell of apoptosis,infrequently erythraeous necrotic cell. PA,CDDP group:The apoptosis rate was(9.70±1.42)%,(7.25±1.07)%.fluorescence microscope it was observed that predominant tumorous cell of virent fluorescence, evident augmented yellowish green cell of apoptosis and erythraeous necrotic cell compared with control group. Quantity of cell apoptosis compared with 5-Fu group.PA with 5-Fu group,PA with CDDP group:The apoptosis rate was(14.74±1.39)%,(20.51±1.61)%. Combined medicines are more efficient enhancement to Hep-2 cell apoptosis than single medicine, acridine orange staining disclose it ,that more cell apoptosis and conferted trachychromatic in massis yellowishgreen cell of apoptosis. Statistics demonstrates:combined medicines, single medicine apoptosis rate were both increased compared with control group (P<0.05). Combined medicines apoptosis rate was increased compared with single medicine(P<0.05). 2.The result of immunohistochemistry: Bax was showed it, that negative in control group, positive express were single scattering amber-coloured grana in cytoplasm and cell membrane in single medicine(PA, 5-Fu ,CDDP group);the overexpressed was amber-coloured focal focusling in cytoplasm and cell membrane in combined medicines group(PA with CDDP,PA with 5-Fu). Statistics demonstrates:expression of Bax protein was significantly potentiating in combined medicines, single medicine group(PA,CDDP group) compared with control group(P<0.05). Expression of Bax protein was significantly potentiating in combined medicines group compared with single medicine group (P<0.05).Conclusions: 1,Combined medicines are more efficient enhancement to Hep2 cell apoptosis than single medicine. 2,Combined medicines are more efficient for apoptosis of Hep2 cell than single medicine.3,That enhancing apoptosis of Hep2 cell by treatment of combinig the mechanism of Phenylacetic acid with chemotherapeuatic medicine perhaps have some relations with the up-regulation of the expression of proapoptotic Bax .
