An Experimental Study On The Effects Of Stem Cells Of GFP Transgenic Mices And Anti-epileptic Drug On Epileptic Rats

The research parts of this thesis include "The effect of implantin stem cells on epileptic rats and expressing of Bax protein and Bcl-2 proteiry " (part I) and "The effects of anti-epilepitic drugs on behaviors electroencephalogram and the expressing of Bax and Bcl-2 of The epilepitic rats induced by penicillin" (part II) .Part I: The effect of implantin stem cells on epileptic rats and expressing of Bax protein and Bcl-2 proteiryObjective: There are no report about implanting stem cells to treat epilepsy in domestic and international. To test the effect of transplanting neural stem cells and bone stromal stem cells to the hippocampus of epileptic rats, We observed and explored to the survival and migration of the stem cells, the integration with and repair of the brain tissues, and to test the effect of the stem cells of the apoptosis in the epileptic rats. To provide some experimental bases that transplanting stem cells can treat epilepsy.Methods The epileptic rat models were produced after penicillin was injected into the belly of the rats. The rats were divided into normal control group(n=8),group of the epilepsy without treatment (n=8), group of the epilepsy with Sodium Chloride (n=24), group of the epilepsy with neural stem cells (n=24), group of the epileptic with bone marrow stromal stem cell (n=24).Neural stem cells (NSCs) and bone marrow stromal stem cells (BMSCs) of transgenic mice with green fluorescent protein (GFP) were isolated and cultured. The suspension of NSCs and the suspension of BMSCs were respectively grafted into the right hippocampus of the rats with the stereotaxic apparatus.Then the rats were respectively perfused at 1week, 2weeks, 4weeks after grafting.The survival and immigration of the neural stem cells and bone marrow stromal stem cells were observed under the fluorescence microscope.The shapes of the apoptosis...
