Effects Of Therapies NSCs Transplantation Combined With DNA Vaccine Of NGIs On The Motor Functional Recovery Following Spinal Cord Injury

[Objective] To investigate the effects of recombinant DNA vaccine encoding multiple domains related to inhibition of neurite outgrowth combined with transplant of neural stem cells on the motor functional recovery following transected spinal cord in rats.[Methods] SD female rats, weighting from 180g to 200g, were randomly divided into 4 groups: 1.spinal cord transection group in which the spinal cord was transected at T9-10 completely; 2.spinal cord transaction combined with treatment of recombinant DNA vaccine related neurite growth inhibitors; 3.spinal cord transection combined with treatment of transplanted neural stem cells; 4.spinal cord transection combined with the recombinant DNA vaccine and transplant of neural stem cells. Behavioral evaluation was performed by BBB score after transplantation 1 week, 2 weeks, 3 weeks, 4 weeks. The survival, migration and differentiations of transplanted neural stem cells are investigated by sections of spinal cords after 4 weeks post operation.[Result] The BBB scores of animals in all groups were increased from 1 week post operation to 4 weeks, especially the score of 4 weeks was significantly increasing than 2 weeks (P<0.05). In different groups, the scores of animals accepting therapeutic alliance were higher than other groups at 3 weeks after transplant. At 4 weeks, motor function recovered more evidently in animals accepting therapeutic alliance or neural stem cell transplant than operation group and animals injected DNA vaccine (P<0.05). Histological observation: In fluorescence microscope, transplanted neural stem cells around the injured regions survived well and migrated to peripheral normal tissue of injured spinal cord, especially in group receiving therapeutic alliance (P<0.05). In light microscope, some transplanted cells were differentiated to become neurogliocyte-like or neuron-like cells.[Conclusion] NSCs transplant conduced to functional recovery following transected spinal cord of rats. The application of recombinant DNA vaccine related to neurite growth inhibitors didn't significantly affect functional recovery. The treatment combined with cell transplant and DNA vaccine injection can promote the recovery of hind limb function more effectively than single therapy of cell transplant or DNA vaccine only.
