Objective: To study the frequency of HLA-A*02 alleles in HLA-A*02 positive Xuanwen human lung cancinoma and acquire the distribution of HLA-A*02 allele subtypes.Methods:48 patients residing in Xuanwei country, with lung carcinoma diagnosised between July 2005 and March 2007.These patients were selected randomly from department of radiotherapy and thoracic surgery. 40 normal healthy individuals were randomly selected form donors who settle in Xuanwei. Allele-level HLA-A typing was accomplished primary by PCR with sequence-specific primers(PCR-SSP) to type 48 cases and 40 donors. Then, High resolution typing methods of HLA-A*02 PCR-SSP were employed to type the HLA-A*02 positive case. Case-control study was used to study the association between case subject and control subject. To investigate the distributions of HLA-A*02 alleles in Xuanwei human lung carcinoma.Result: In case subject, gene frequency of allele A*02 was significantly higher compared to the control subject(P<0.05,OR=2.432,95%CI=1.018~5.806) ,other A alleles were not significantly different. Four different HLA-A*02 alleles(A*0201,0203,0206 and 0207) were identified in case subject, of which A*0207 was the predominant allele. When comparing the frequency of A*02 alleles in lung carcinoma and Han populations in south China, the A*02 alleles were not significantly different between them. Seven A*02 homozygote were identified in Xuanwei human lung carcinoma at the low-and high-resolution levels. The predominant allele of homozygote was A*0207 43% (3/7) .Conclusion: HLA-A*02 may be associated with increased susceptibility to lung carcinoma. In the Xuanwei lung carcinoma, HLA-A*0207 was the predominant allele. The distributions of A*02 allele subtypes belongs to the Han population in south China.
