Mammary gland hyperplasia is one of the most common mammary diseases during the Pregnant and Perinatal Period. In recent years, a lot of researches have been done on acupuncture which played a more and more important role in the treatment of mammary gland hyperplasia. But there is lack of experimental evidence on the effect of acupuncture on mammary gland hyperplasia. Based on the modern research and the guidance of ancient literatures, this research explored the effect of acupuncture on the hormone levels of hypophysis-ovary sex gland axis in rats with mammary gland hyperplasia by observing the level of P, E2, PRL, LH, FSH before and after treatment.The breast is the target organ to the effect of sex hormones. The occurrence of Mammary gland hyperplasia is due to the deficient secretion of LH and E2 in preovulatory phase, and the increase of E2, deficient secretion of P in luteal phase. The increase of PRL inhibits the secretion of P and promotes the synthesize of E2 in the luteal phase and causes disproportion of E2/P. The research induced mammary gland hyperplasia in rat with estrogen. The rats were randomly assigned into normal group, model group, tamoxifen group and acupuncture group. The effects of acupuncture and tamoxifen on the 5 kinds of sex hormones (P, E2, PRL, LH, and FSH) were observed after the treatment. The 3rd pairs of mammary tissues were taken for pathological section to be observed under light microscope to compare the numbers of mammary acinus, the diameter of acinus cavea and duct.The following results have been found in the experiment:1. The height of nipples increase by different levels in each group. The increase in the model group was most significant and had remarkable difference comparing with normal group (P<0.01); The nipple heights in the tamoxifen group and acupuncture group are obviously lower than those in the model group. The nipple height of tamoxifen group had significant difference comparing with model group (P<0.01), The above two groups has remarkable difference comparing with normal group.2. The array of the mammary gland duct epithelia in the normal group is regular. There is no dilatation in the duct cavities and acinus' s cavities. The number of lobule and acini is rare. In model group, irregular array of the mammary gland duct epithelia are found with obvious hyperplasia of the epithelia, dilatation of the duct cavities and acinus' s cavities, dropping epithelia cells and secretion, increased lobules and acini, and obvious hyperplasia of the mesenchyme fiber connective tissues, which indicates the successful model creations. The manifestation in tamoxifen and acupuncture group are similar as the following: normal array of the mammary gland duct epithelia, less dilatation of the duct cavities than that in the model group; obvious reduce of the size of the lobules and lobule acini; insignificant hyperplasia of the fibre connective tissues, especially in the tamoxifen group.3. After the treatment, the level of serum E2, P, PRL, LH, FSH in each group were observed. The levels of E2(P<0.05),PRL (P<0.01), FSH(P<0.01) in model group were significantly higher than those in the model group; The level of LH (P<0.01),P (P<0.05) were significantly lower than those in the normal group. After treatment, the levels of E2, PRL, and FSH were lower than those in the model group. The level of LH and P has been increased differently.In summary, the following conclusions have been drawn: 1. acupuncture can improve the mammary structure in rats with mammary gland hyperplasia, re-array the irregular duct epithelia, reduce the expanding of the acinus cavae. 2. Acupuncture may inhibit the mammary gland hyperplasia by adjusting the sex hormones level of rats and repairing the lesion of the tissues. In addition, acupuncture has a beneficial bidirectional regulation to the sex hormone levels and wonderful prospects in its clinical research.
