Objective:To study the main HPV infection types and genotyping of the CINpatients in Qingdao area. To study the prevalent HPV types and the single and multipleHPV infection in different grade of CIN.To assess the validity of HPV DNA detectionand typing with PCR from the paraffin-embedded tissues.Methods:Type-specific PCRwas used to detect HPV and genotyping of cervical intraepithelial neoplasia (CIN)from the paraffin-embedded tissues in Qingdao area.Result:(1)There were 94 sampleswere screened for the presence of HPV DNA form 114 patients in Qingdao area, thepositivity rates of total HPV was 82.46%.The infection rates of the four HPV types aredifferent((X~2=42.632, P<0.001).the prevalence of HPV types from highest to lowestorder were as follows: HPV 16 (54.39%), HPV 58 (35.09%), HPV 18 (29.82%),HPV 33 (14.04%). (2). The positivity rates of total HPV in different grade of CINwere different (X~2= 25.762, P<0.001). the positivity rates of total HPV in CINⅠ,CINⅡ, CINⅢwere53.85%; 72.73%; 96.97%respectively. Among these, CINⅢwas higher than the other two groups,although CINⅡis higher than CINⅠ,but there isno significant difference (P>0.05). (3). The prevalence of different HPV types fromhighest to lowest order in different grade CIN were as follows: HPV 58; HPV 16;HPV 18; HPV 33 for CINⅠ;. HPV 16; HPV 18; HPV 58; HPV 33 for CINⅡ; HPV16; HPV 58; HPV 18; HPV 33 for CINⅢ.(4). The single and double HPV infectionwere more common than other HPV infection in CIN, triple and quadruple HPVinfection were also detected. The multiple HPV infection rate was different in differentgrade of CIN (X~2= 14.419, P<0.01),among these the double infection was the mostcommon. (5). In this study,we extracted the high quality DNA from theparaffin-embedded tissues. This supplied us a valid method to studyCIN.Conclusion:(1) The prevalence of HPV types of CIN from highest to lowest orderin Qingdao area were as follows: HPV 16, HPV 58, HPV 18, HPV 33. (2). The positivity rates of total HPV in different grade of CIN were different, HPV positiverates increased with the seriousness of cervical lesions. The prevalence of differentHPV types in different grade of CIN were different. All of these give evidences to thetreatment and screening of the cervical lesions.(3).The single infection was the mostcommon infection in CIN, at the same time ,there also existed multiple infection. Themultiple HPV infection rate was different in different grade of CIN.(4)The method ofextracting DNA for PCR from the paraffin-embedded tissues was valid.
