Objective TO observe the expression of Toll like receptor 4 (TLR4), glialfibrillary acidic protein(GFAP), myelin basic protein (MBP) and platelet-derivedgrowth factor receptor alpha (PDGFR-α) in The Brain of The premature Rats withintrauterine infection The aim of the study is to explore the mechanisms of immunologybetween intrauterine infection and white matter damage (WMD), so as to provide theexperimental base for finding clinical diagnosis and treatment of WMD.Methods Pregnant rats were randomly divided into two groups:infectiongroup and control group. The infection group was established by intraperitonealinjection of endotoxin in pregnant rats when gestation was 70% complete (15days). The control group was treated with normal saline. The fetal rats were killed aftermaternal endotoxin administration2,4,12,24and72hours respectively. The pathologicalchanges in placenta and in fetal rat brain were determined by hematoxylin andeosin(HE) staining. Using fluorescence RT-PCR to examine the expression of TLR4in fetal rat brains. Immunohistochemical method was used to detect the expression ofGFAP, MBP and PDGFR-α.Results After maternal endotoxin administration, infiltration of neutrophil inthe placenta, weak staining of cerebral white matter and focal rarefaction in the fetalrat brains were observed. Expression of TLR4 mRNA in the fetal brain was decreasedin 2h and 4h after the administration of LPS. In the endotoxin-treated group, positivestaining of MBP and PDGFR-αin the fetal rat brains was much less and weaker in12h,24h,72h; the increased expression of GFAP was observed in 12h,24h,72h.Conclusions Endotoxin can be used to establish intrauterine infection modelsand the infection may cause the lesion of the white matter. TLR4 participate in thecourse of the injury introduced by LPS.
