Objective To transfect the pcDNA4C-TNF-βcDNA into HepG2 by cationieliposome mediated method and detect the expression of TNF-βand Survivin in transferedHepG2 cell. Methods The pcDNA4C-TNF-βcDNA extracted from E.coli JM-109, wastransfected into HepG2 by cationie liposome mediated method. HepG2 cell andsupernatants was collected after transfection of 12h, 24h, 48h, 72h and 5d respectively.The collected cell was disrupted. TNF-βprotein was detected in the supernatants byELISA. Survivin protein was detected in the lysate of the cell disruotion. Standard curveswere obtioned from the set of samples assayed. Results In transfection group, theexpression, of TNF-βwas identified by ELISA(r=0.991, p<0.001 with non-transfectiongroup). The content of TNF-βprotein was the higher at 24h and the lower at 5d. Innon-transfection group, the expression of Survivin was succesfully identified byELISA(r=0.985, p<0.001 with transfection group). The content of Survivin protein washigher at 24h and lower at 5d. Conclusion The pcDNA4C-TNF-βwas effectivelytransfected in HepG2 mediated by liposome. Sustained expression was obtained. Innon-transfection group, the expression of Survivin was succesfully identified. Theexpression of Survivin decreased obviously after transfection.
