Objective To transfect the LIGHT and IFN-γinto HepG2 cells mediated by cationie liposome and detect cell apoptosis and the expression of Survivin and Caspase-3 in transfered HepG2 cells. Methods The pcDNA4C-LIGHT-cDNA and pcDNA4/ HisMax-hIFN-γextracted from E.coli JM-109, were transfected into HepG2 cells mediated by cationie liposome. The HepG2 cells was collected after transfection of 12h, 24h, 48h respectively. The cell apoptosis and the expression of Survivin and Caspase-3 was detected by flow cytometer(FCM). Results The cell apoptosis and expression of caspase-3 increased by degrees among control group, LIGHT-mediated group and cotransfection group, the expression of Survivin decreased by degrees. Conclusion LIGHT had an effection on inducing the apoptosis of HepG2 cells by regulating the expression of caspase-3 and Survivin, probably. IFN-γcan sensitize its effection significantlly.
