Objective: To investigate the expression of ATF3 protein in human prostatic carcinoma , benigh prostatic hyperplasia as well as normal prostate tissues. To explore the role ATF3 played in the oncogenesis,development, metastasis of human prostatic carcinoma. To determine the relationship between ATF3 expression and the clinicopathological features of human prostatic carcinoma.Methods: The expression of ATF3 protein was detected in human prostatic carcinoma , benigh prostatic hyperplasia as well as normal prostate tissues using immunohistochemistry and Western Blot . The ATF3 expression was compared in different tissues. The relationship between ATF3 expression and relatied clinicopathological features was statistically analyzed.Results: The immunohistochemistry detection of ATF3 protein in paraffin slices indicates that the positive rate of ATF3 in prostatic carcinoma(58.70%)was significantly higher than that in benign prostatic hyperplasia(13.04%)and normal prostate (0%)(P<0.01). The cases were classified according to Whitmore-Jewett staging system and graded according to Gleason grading system. The expression of ATF3 protein was associated with pathological grade and clinical stage but have no relationship with preoperative prostate-specific antigen level. The expression of ATF3 protein in prostatic carcinoma in C+D stage was higher than that in A+B stage. The expression of ATF3 protein in prostatic carcinoma with metastasis was higher than that without metastasis. The Western Blot reveal that the expression of ATF3 protein was present in all three groups and the amount of ATF3 protein in prostatic carcinoma tissues was higher than that in benigh prostatic hyperplasia or normal prostate tissues singnificantly.Conclusions: The expression of ATF3 in prostatic carcinoma was significantly higher than that in benigh prostatic hyperplasia or normal prostate. The expression of ATF3 protein was associated with pathological grade ,clinical stage as well as metastisis but have no relationship with preoperative prostate-specific antigen level .It indicate that ATF3 may play a role in the oncogenesis,development, invasion and metastasis of prostatic carcinoma. Detection of ATF3 protein may reflect the malignancy and prognosis of human prostatic carcinoma. Further study to the molecular mechanism of the ATF3 induction and related signal transduction pathways may reveal the pathogenesis of prostatic carcinoma.
