Objective: To investigate the enhancement of mouse nerve growth factor continuously in nerve regeneration chamber on the motor end plate and muscle.Method: Twenty-four Sprague-Dawley rats were used in this study. Their two sciatic nerves were transected to form a 3 mm gap and bridged with silicone tube. On randomly principle, one sciatic nerve was treated with mouse nerve growth factor 22.5Au in nerve regeneration chamber and continuously in it every 3 days as the experiment group, and the other were treated with nothing in the other side as the control group. At the time of 2,4,6,8 weeks after operation, the motor end plate and muscle was evaluated with gross observation, electrophysiological test, the wet muscle weight, the cross section area of muscular, the changes of calcitonin gene-related peptide(CGRP)in skeletal muscle.Result: The motor nerve conductive velocity (NCV) of experiment group was significant faster than that of control group (p<0.05). The amplitude of compound muscle action potential (MAP) was significant bigger (p<0.05). The wet muscle weight of experiment group was significant much more than that of control group, and the cross section area of muscular was significant bigger(p<0.05). The level of CGRP in skeletal muscle of experiment group was significant much more than that of control group.The improvement of NCV, the MAP, the wet muscle weight, the cross section area of muscular, the level of CGRP in skeletal muscle in both experiment group and control group were time-dependent, the latter, and the better(p<0.05).Conclusion:①Continuously local administration of nerve growth factor takes effect on the motor end olate and muscle regeneration and repair, so takes effect on promoting constitution and function of peripheral nerve.②The nerve regeneration chamber in the silicone tube plays an important role of mechanical support and guiding action in bridging nerve defect and keep NGF effective density. The nerve provides chemotaxis for nerve regeneration, so take effect on target organ.③Nerve growth factor continuously in nerve regeneration chamber is a effective method.
