Objective: This study is designed to measure the contents of OPNand TNF-αin the synovia fluid with knee osteoarthritis (OA), in order todemonstrate the relationship of OPN and TNF-αto OA, and study thereeffect on OA pathogenesis and there clinical significance.Methods: OPN and TNF-αwere measured by enzyme-linkedimmunosorbent assay(ELISA) concurrently in SF samples from 50patients with knee OA and from 6 controls. Statistical analysis were madeto demonstrate the relation between each other.Results: Joint fluid contents of OPN and TNF-αin OA group weremarkedly higher than those in control group with a significant difference(p<0.05). The contents of OPN showed no significant difference betweenmild group and moderate grup(P>0.05), and there were significantdifference between severe group and others(p<0.05). The contents ofTNF-αshowed no significant difference in three groups of OA(P>0.05).The contents of OPN showed a direct correlation with the severity ofOA, and it also showed a direct correlation with the contents ofTNF-α. There was not correlation between TNF-αlevels and the severity of OA.Conclusion:1. OPN and TNF-αplay important role in OA pathogenesis.2. It showed that the contents increasing of TNF-αmight induce thatof OPN and the two factors might play synergistic action in OApathogenesis.3. Both OPN and TNF-αcan be regarded as markers for earlydiagnosis of OA, and only OPN can be the marker to show the severity ofOA.
