Objective : To study the effect of thalidomide (Thd) used on the growth and angiogenesis of human breast cancer transplanted subcutaneously in nude mice.Methods : human breast cancer model transplanted subcutaneously in nude mice was established, and divided into 4 groups: control group, Thd group, and Thd + CTX group, CTX group . NS (0. 2ml) ,Thalidomide (200 mg/ kg) , CTX (100mg/kg mg/ kg) ,or Thalidomide (200 mg/ kg) plus CTX (100mg/kg mg/ kg) was peritoneally injected respectively for each group from the second day of building model. The mice were sacrificed on the 28th day. The weight of tumor , latent period and percent of tumor development were observed. Immunochemistry staining for factor VID was performed and MVD was calculated.Results: 1.The tumor weight and volume of treated group were significantly lower than that of control group except Thd group , these is statistical difference (P<0.05 ) . The tumor weight and volume of the Thd combined with CTX group were lower than that of Thd group and CTX group , these is statistical difference (P<0.05) .2. Immunochemistry staining showed irregular angiogenesis without obvious lumens mostly scattered around edge of tumor. MVD in Thd group and Thd +CTX group was significantly smaller than that in control group except Thd group, these is statistical difference (P<0.05) .No statistical difference was found in tumor growth between thalidomide group and control group(P>0.05) .Conclusions: 1. Thalidomide has the tendency to inhibit the growth of human breast cancer MBA-MD-231 transplanted subcutaneously in nude mice. It is necessary to increase the sample and extend the time of observation.2. Thalidomide can significantly inhibit the angiogenesis of human breast cancer MBA-MD-231 transplanted subcutaneously in nude mice.3. CTX can inhibit the growth of human breast cancer MBA-MD-231 transplanted subcutaneously in nude mice and the effects of CTX anti-tumor was strongly amplified when thalidomide was added .
