ObjectiveTo study the effect of thalidomide usedon ovarian cancer cell subcutaneous implanted tumor growth inhibition.Methods1,20 ovarian cancer modle ubcutaneous implantted in nude mice was established and randomly divided into 4 groups, n=5:(1)thalidomide group:200mg.kg-1d-1,0.2ml, ip.(2)DDP(cisplatin)group:0.2mg.kg-1d-1,0.2ml, ip.(3)Combined treatment group:both Thd and DDP,0.2ml, ip.(4)control group:saline solution,0.2ml, ip.2,After the success of the implant, the MTD(mean tumor diameters) 0.8cm, administered two weeks, twice a week, a total of 4 injection. The groups weight observed in each group,MTD changes, and the actions of nude mice, the response to external stimulation, food, the surface of the skin tumor changes and measured tumor inhibition rate.3,Animals were killed 2 weeks after administration, the MVD (tumor microvessel density) and apoptosis index(AI) were detected by the immunohistochemisty or TUNEL in tumors tissue.Results 1,Compared with the control group, growth of tumor was significantly reduced in mice treated with thalidomide, DDP and combined treatment. (inhibition rate 57.47%, 42.18%,71.37%)(P<0.05).2,Control group MVD16±2.92, thalidomide group MVD5.8±0.84, DDPgroup MVD16.20±1.92, Combined treatment group MVD2.8±0.84. In thalidomide group and combination group MVD was significantly inhibited(P<0.05).3,Control group AI7.202±0.44, thalidomide group AI12.586±0.69, DDPgroup AI10.30±0.72, Combined treatment group AI17.998±0.61. In thalidomide group, DDPgroup and combination group AI was significantly higher (P<0.05). Combination group AI was significantly highest(.P<0.05).Conclusions1,Thalidomide can inhibit the growth of human ovarian cancer subcutaneous implanted in nude mice. Thalidomide can induce apoptosis in ovarian cancer by inhibiting tumor angiogenesis.2,In combination with DDP, thalidomide might prove the antitumor drug therapy.
