Objective:To study the histological effect of ultrashort wave therapy self-repair rupture of medial collateral ligament in rabbit's knee.Methods:20 adult rabbits were randomly divided into control group(intersect of medial collateral ligament on operation )and ultrashort wave therapy group (Treat by ultrashort wave therapy 24 hours later since intersecting their medial collateral ligament), one time a day, gentle heat, 10~15 minutes, ten times for course of treatment) (each n=10). Samples were collected 7 weeks later since operation. Healing of the ligament was observed by macroscopic and HE staining. Type I and III collagen were measured grey after IH staining.Results:1. Macroscopic observation: control group, the surface of broken ends of MCL was repaired fairly rough and dim, shrink and thinning compare with normal ligament, adhered to surrounding tissue. In the ultrashort wave therapy group, the surface of the broken ends of MCL was repaired smooth and glossy, showed white color and thicken compare with normal ligament, clear limit with surrounding tissue, without adherence.2. HE staining: control group, there were a great quantity of fibroblast-like cell proliferation, arranging no regularity, the most cellular nucleus were round and oval; collagen fiber was acidophilia, schillerization, arranged disorderly, normal collagen fiber bundle to disappeared. To the ultrashort wave therapy group, the number of fibroblast-like cell decreased compare with control group, collocation obviously regularity, the most cellular nucleus were rhabditiform and spindle, the round and elliptical cells obviously decrease; collagen fiber arranged regularity,collagen fiber bundle emerged.3. Immunohistochemistry: type I collagen were positive staining, grey of control group and ultrashort wave therapy group was (27.83±3.10) and (23.35±3.80) respectively. Type III collagen were positive staining, grey in control group and ultrashort wave therapy group was (80.91±4.82) and (76.12±5.19) respectively. Grey in ultrashort wave therapy was significantly less than that in control group(P<0.05). Grey measured at each point is corresponding Type I and III collagen content of that position, the grey is in inverse proportion to content.Conclusions:1. Ultrashort wave therapy promoted self-repairing rupture of medial collateral ligament in rabbit's knee.2. Rupture of medial collateral ligament can self-repair.
