Objective: To investigate the expression of E-cadherin, Dysadherin and lymphatic vessel density as well as the correlation between the expression of the three factors and clinical significance in laryngeal carcinoma in order to find the carcinomal metastatic symbols which might predict the progression, metastasia and prognosis of the tumor.Methods: Western-blot analysis was performed on tumor core region, tumor transition region, tumor free region of 40 specimens of fresh LSCC and 10 specimens of normal mucosa using anti-E-cadherin, Dysadherin antiserums. Immunohistochemistry was performed to quantitate lymphatic vessels with lymphatic endothelial marker D2-40 on 40 specimens of the LSCC and 10 specimens of normal mucosa.Results: The expression of E-cadherin protein level from tumor core region, tumor transition region and tumor free region of LSCC appeared to increase progressively, The expression of Dysadherin protein level from tumor core region, tumor transition region and tumor free region of LSCC appeared to decrease progressively; There was negtively correlation between expression of E-cadherin and expression of Dysadherin (P<0.01) ; The expression of Dysadherin might be related to the clinical staging(P<0.05), lymph node metastasis(P<0.01), clinical classification (P<0.01) and the degree of differentiation (P<0.05); LVD might be related to the clinical staging (P<0.05) , lymph node metastasis (P<0.05) , and clinical classification (P<0.01) ,but no related to the degree of differentiation (P>0.05), Expression of Dysadherin correlated positively with LVD (P<0.01) ; Expression of E-cadherin had no correlation with LVD (P>0.05) .Conclusion: There was negtively correlation between expression of E-cadherin and Dysadherin; The overexpression of Dysadherin maybe promoted lymphangiogenesis and highly associated with the growth and metastasis of LSCC,and may be of prognostic value; The LVD of peritumor was highly associated with the growth and metastasis of LSCC.
