Laryngeal cancer is one of the most common malignant tumour in Head and Neck cancer,and 96%-98% of laryngeal carcinoma is Squamous Cell Carcinoma of the larynx(laryngeal squamous cell carcinoma,LSCC).The most recent studies have shown the downward trend in the overall survival rate of laryngeal cancer,as cancer cell invasion and lymph node metastasis are key factors affecting the prognosis of laryngeal carcinoma.Further studies of the molecular mechanisms,which related to the invasion and metastasis of laryngeal cancer,can will providing the necessary basisfor for prognosis and individualized treatment strategies of patients with laryngeal cancerTumor invasion and metastasis is a dynamic and complex process,is the result of many protein molecules involved in multiple steps.Ezrin,as the connection of membrane and cytoskeleton proteins,showed high expression in many malignant tumors,play an important role in tumor invasion and metastasis,and is considered to be a key regulator of tumor metastasis.RhoA mediated many signal transduction pathways involved in regulating cell activity as the switche of signal path,RhoA may via activating ROCK-1 phosphorylates Ezrin,strengthen its links with the cytoskeleton,studies in recent years have shown that expression of RhoA is closely related to tumor development.This study aims to analyse the expression of Ezrin and RhoA in laryngeal carcinoma and the correlation with the clinical informations,to reveal the mechanisms of invasion and metastasis in laryngeal carcinoma.??The expression of RhoA and Ezrin in fresh tissueApplication of Immunohistochemistry(IHC),Western blot(WB)and Quantitative reverse transcription polymerase chain reaction(qRT-PCR)in 30 cases of fresh laryngeal squamous cell carcinoma and 30 cases of normal laryngeal mucosa show the expression and Clinical pathological features of RhoA and Ezrin.Finally,make a statistical analysis of the above information by using SPSS 22.0 software,while P < 0.05 has significant differences.Immunohistochemistry shows the expression of RhoA and Ezrin(60% and 70%)in LSCC were significantly higher than normal laryngeal mucosa(13.3% and 16.7%)(P<0.05);Western blot results showed RhoA and Ezrin in the LSCC tissue weight were significantly higher than that of normal laryngeal mucosa in the level of protein Molecular Weight;qRT-PCR results shows the RhoA and Ezrin in the LSCC were higher than normal laryngeal mucosa in the level of mRNA expression.Their expression is closely related to clinical stages and metastasis(P<0.05),not related to age,gender,smoking status or differentiation(P>0.05).Kappa results indicated the consistency between the expression of Ezrin and RhoA(Kappa=0.638).??Restrospectively analyzed the expression of RhoA and Ezrin in LSCC and normal laryngeal mucosaApplication of Immunohistochemistry(IHC)in 68 cases of laryngeal squamous cell carcinoma and 35 cases of normal laryngeal mucosa show the expression and clinical pathological features of RhoA and Ezrin.Results showed the expression of RhoA and Ezrin(80.9% and 82.4%)in LSCC were significantly higher than normal laryngeal mucosa(28.5% and 25.7%)(P < 0.05);Their expression is closely related to clinical stage and metastasis(P<0.05),not related to age,gender,smoking status or differentiation(P > 0.05).Consistency Check indicated the consistency between the expression of Ezrin and RhoA(Kappa=0.363).It is the same with expression of Ezrin and RhoA in fresh tissue,make the results more reliability.In summary,RhoA and Ezrin shows higher expression in the LSCC than in normal laryngeal mucosa,and the expression closely related to TNM stage,and their expressions are consistent.Indicate that they may be common participation in the invasion and metastasis of laryngeal cancer.Immunohistochemistry shows RhoA expressed in the cytoplasm and cell membrane,Ezrin is mainly distributed in the cell membrane.Suggested Ezrin may pass through binding to cell membrane protein to connected actin with the cell membrane,thus changes in cytoskeletal structure and regulates cell adhesion,to participate in tumor cell invasion and metastasis process.RhoA may as the activator of Ezrin in the process of tumor invasion and metastasis.Provide new evidence for early diagnosis ?prognosis and treatment of laryngeal cancer.
