Objective The aim of this study was to detect the expression of Ezrin and E-cadherin in human advanced gastric carcinoma and their metastatic lymph nodes on mRNA and protein level,discuss the relationship between The expression of Ezrin E-cadherin and the invasiveness and metastasis of gastric carcinoma,so that we can in serch of a effective Method in Molecular Biology for early gastric tumour diagnosis and treatment.Methods: This study selected 45 cases of advanced gastric carcinoma (31 cases have metastatic lymph nodes) as the experimental group and 25 cases of benign gastric mucosal lesion as the control group,assumed three methods of Immunohistochemistry Western blot and RT-PCR,so that We could approach the connection between the expression of Ezrin E-cadherin and the clinicopathologic characteristic of gastric carcinoma ,detect the mRNA and protein expressive levels of Ezrin and E-cadherin in corresponding tissues,then research the correlation between Ezrin and E-cadherin,and analyze the pertinence between The expression of Ezrin E-cadherin and the invasiveness and metastasis of gastric carcinomaResults (1) Immunohistochemical results showed that in 45 cases of primary gastric carcinoma tissues, the overexpression rate of Ezrin was 73.3% (33/45), the abnormal expression rate of E-cadherin was 71.1%(32/45).In 31 cases of metastatic lymph nodes, the overexpression rate of Ezrin was 96.8% (30-31), the abnormal expression rate of E-cadherin was 93.5% (29/31).The overexpression rate of Ezrin in metastatic lymph nodes was significantly higher than in the primary niduses (P = 0.008), analogously ,the abnormal expression rate of E-cadherin in metastatic lymph nodes was also prominently higher than in the primary niduses(P = 0.016).In addition,the over-expression rate of Ezrin in stomach primary foci was closely related to the pathologic grade (P =0.014),but not related to histological type of carcinoma(P = 0.524) or patients gender(P = 0.821) and age(P = 0.725).In stomach primary focal tissues with lymph node metastases, its excessive expression rate was significantly higher than these cancerous tissues without lymph node metastases (P = 0.000).Accordingly,the abnormal expression rate of E-cadherin in stomach primary foci was also closely contact to pathologic stage (P = 0.032) and lymph node metastases(P=.005), but not obviously related to histological type of carcinoma(P = 0.314) or patients gender(P = 0.419) and age(P = 0.165).There was a negative correlation between the expression of Ezrin and E-cadherin by Spearman rank correlation analysis(rs=-0.621,P=0.000).According to the conjointing analysis of Ezrin and E-cadherin We found that the lymph metastasis rate of the group which excessive(abnormal) expression rate of Ezrin and E-cadherin were both higher was significantly higher than the other group with single protein excessive(abnormal) expression or without excessive (abnormal) expression2=4.055 P=0.044 .(2) Western blot results showed that in benign lesions of gastric mucosa , the protein relative strength of Ezrin was 6.0-95.3 54.248025.62436 ,in the primary niduses of gastric carcinoma,the corresponding data was 13.5 -117.4(69.4933 + 29.98342),obviously higher than the former,but the uppermost index was in the metastatic lymph nodes,it was 21.5-135.1 69.4933 29.98342 ,the compare between each group was significantly different (F = 7.226, P = 0.001). On the contrary, the E - cadherin protein relative strength of benign gastric mucosal lesions was 25.4-142.3(88.644 37.0130 , on the higher level than the stomach primary foci,and the data of the latter was 19.1-131.5 70.109 32.8545 ,but the minimum was 10.2-102.5 53.865 28.1045 in the metastatic lymph nodes below the first two,the compare between each group was also significantly different (F = 7.888, P = 0.001). The protein relative strength of Ezrin in the primary gastric niduses having corresponding metastatic lymph nodes 75.9677 28.35094 was taller significantly than in the tissues without corresponding metastatic lymph nodes 55.1571 29.43853 t=2.235 P=0.029 . Conversely, the protein relative strength of E - cadherin in the primary gastric niduses having corresponding metastatic lymph nodes 63.49430.6554 was even significantly lower than in the tissues without corresponding metastatic lymph nodes 86.257 33.4295 t=-2.243P=0.030 .(3)RT-PCR results were in accord with Western blot.The results showed that in benign gastric mucosal lesions, the relative expression value of Ezrin mRNA was 0.75424 0.004876,in the primary niduses of gastric carcinoma,the corresponding data was 0.77764 0.006311,obviously higher than the former,but the uppermost index was in the metastatic lymph nodes,it was 0.78529 0.006861,the compare between each group was significantly different F=187.680 P=0.0000 . Conversely, the values of E-cadherin mRNA of benign gastric mucosal lesions was 0.74072 0.014296, on the higher level than the stomach primary foci which data was 0.730670.014402,but the minimum was 0.71835 0.010268 in the metastatic lymph nodes below the first two,the compare between each group was also different significantly F=20.113,P=0.000 . The relative expression value of Ezrin mRNA in the primary gastric niduses having corresponding metastatic lymph nodes 0.77894 0.006408 was taller significantly than in the primary niduses without corresponding metastatic lymph nodes 0.77479 0.005250t=2.119 P=0.040 . On the contrary, the relative expression value of E - cadherin mRNA in the primary gastric niduses having corresponding metastatic lymph nodes 0.72658 0.13841 was even significantly lower than in the primary niduses without corresponding metastatic lymph nodes0.73971 0.11485 t=-3.096 P=0.003 .Conclusion: The abnormal expression of Ezrin and E - cadherin was universally exist in gastric carcinoma.The expressive level of Ezrin in the stomach primary foci was higher than in the benign lesions of gastric mucosa , however the corresponding data in metastatic lymph nodes was significantly higher than the former two. In the meantime ,the expressive level of E - cadherin in corresponding groups was on the contrary, in primary gastric niduses,it was appeared in decreasing, lacking expression even heterotopic expression,in gastric lymphatic metastases this phenomenon was behaved more outstanding.Thus,the excessive expression of Ezrin and the expressive abatement or deficiency of E-cadherin all promoted the occurrence,development,invasiveness and metastasis of gastric carcinoma.We found that the lymph metastasis rate of the group with concurrent abnormal expression of Ezrin and E-cadherin was significantly higher than the other group with single protein abnormal expression or without abnormal expression.Excessive expression of Ezrin might affect and reduce the normal expression of E - cadherin, and then cause tumorous metastasis,so we think there was a synergistic process between Ezrin and E-cadherin in promoting tumorous progression and metastasis. Anyhow, Conjoint application of Ezrin and E-cadherin may contribute to the clinicopathologic diagnosis and prognosis of gastric carcinoma,moreover,provide important targets for gene therapy and immunotherapy of tumor.
