Objective To investigate the effects of Shenfu injection on the change of liver cell apoptosis and the expression of NF-κB,HSP70 during hepatic ischemia-reperfusion injury in dogs. Try to evaluate the mechanism of the hepatic ischemia-reperfusion injury in dogs and the protect mechanism of Shenfu injection.Methods 24 healthy hybrid canines were randomly divided into 4 groups:Sham group(only dissected the first porta hepatic, but not blocked),Ischemia-reperfusion group(NS),SF treatment group(2ml/kg),SF treatment group(4ml/kg).It was established the canine model of whole hepatic ischemia-reperfusion with blocked hepatoduodenal ligament. According to time,every group was measured the contents at five time points:before blocked, 15min after ischemis,30min,1h,2h after reperfusion. The expression of NF-κB,HSP70 in liver tissue was accessed by quantitative immunohistochemistry. The liver cell apoptosis index was achieved by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL). Morphological and ultrastructural changes of liver tissue observed by transmission electron microscopy.Results Compared with sham group,ischemia-reperfusion group increased the apoptotic index and the expression level of NF-κB,HSP70 in liver tissue(p<0.05).Compared with ischemia-reperfusion group, SF treatment group(4ml/kg) decreased significantly the apoptotic index and the expression level of NF-κB.However the expression level of HSP70 promoted significantly(p<0.05). Transmission electron microscopy confirmed that the pathological changes of liver tissue were more slight in SF treatment group(4ml/kg) than in ischemia-reperfusion group.Conclusion SF can lessen evidently the hepatic ischemia-reperfusion injury in dogs. It can be explained by decreasing liver cell apoptosis and NF-κB expression,increasing HSP70 expression. And the protective function of SF treatment group(4ml/kg) was more reliable than SF treatment group(2ml/kg).
