Objective To observe the effects of shenfu injection(SF)the content of c-TnT,HSP70 and NF-κB in the canine model of myocardial ischemia reperfusion injury(MIRI).To investigate the possible mechanism of action about SF's anti-MIRI.Methods 20 adult healthy cross bred canines were randomized into four grouds(n=5):sham operated group,control group,SF low dosage group(2ml/kg)and SF high dosage group(4ml/kg).Canines in all groups were injected SF or NS before the operation under the same condition.The model of MIRI was established by ligating left anterior descending coronary artery.The contents of c-TnT in serum were measured at five time points:Pre-drug,ligation,30 rain after ligation,1 hour and 2 hours after referfusion respectively by radio-immunity.The expression of HSP70 and NF-κB in the cardiac muscle tissue were measured at 2 hours after referfusion by immunohistochemistry.Results compared with the sham operated group,the contents of c-TnT,HSP70 and NF-κB of the others all heightened significantly(P<0.05).It was illustrated the success of establishing model.Compared with the control group,SF high and low dosage groups' levels of c-TnT reduced,and the expression of HSP70 strengthened,and the expression of NF-κB lowered (P<0.05).Conclusion SF injection has good protective effects on MIRI,the mechanism of which relates to its function of decreasing the levels of c-TnT,and strengthened the expression of HSP70,and lowered the activity of NF-κB. |
