Objective To observe the expressions of TNF-αand NF-κB in brain tissue following hindlimb ischemia/reperfusion in diabetic rats and the effect of propofol and to investigate the mechanism of action of cytokine and nuclear factor in normal rats and diabetic rats following hindlimb ischemia/reperfusion and the mechanism of protective effects of propofol.Methods Adult male Sprague-Dawley rats weighing 220-250g were randomly assigned to normal rats and diabetic rats.The normal rats were randomly divided into three groups: sham group(ⅠA,n=6),hindlimb ischemia/reperfusion group(ⅠB,n=6),hindlimb ischemia/reperfusion and propofol group(ⅠC,n=6);Diabetic rats were randomly divided into four groups:control group(groupⅡA,n=6),hindlimb ischemia/reperfusion group(ⅡB,n=6), propofol group(ⅡC,n=6),hindlimb ischemia/reperfusion and propofol group(ⅡD,n=6).The diabetic rats induced by streptozotocin were bred for 60 days,the diabetic model was confirmed by measuring the blood glucose(≥13.5mmol/1).Using femoral artery clamped model,the blood stream were reperfused for fours hours after ischemia for three hours.Expressions of INF-αand NF-κB in brain tissue were detected by immunohistochemical and semi-quantitated by image analysis. Expression of TNF-αwere quantitated by enzyme-labeled immunosorbent assay(ELISA).Result①Hindlimb ischemia/reperfusion caused brain injury.The content of TNF-αand NF-κB in brain tissue were significantly higher in groupⅠB than those in groupⅠA.②The content of TNF-αand NF-κB in brain tissue were significantly higher in groupⅡA than those in groupⅠA.③The content of TNF-αand NF-κB in brain tissue were significantly higher in groupⅡB than those in groupⅠA,ⅠB andⅡA.(4)The content of TNF-αand NF-κB in brain tissue were significantly reduced after intervention of propofol.Conclusion①The content of NF-κB and TNF-αincreased in brain tissue of diabetic rats, hindlimb ischemia/reperfusion rats and diabetic rats following hindlimb ischemia/reperfusion, and which was obvious in the last.Diabetes mellitus aggravated brain injury after hindlimb ischemia/reperfusion.②Propofol degraded the content of NF-κB and TNF-αin brain tissue of diabetic rats and hindlimb ischemia/reperfusion rats.Propofol with anti-inflammatory and anti-oxygen role has protective effects to the brain injury induced by hindlimb ischemia /reperfusion in diabetic rats.
