Objective:To approach the protection of ischemic preconditioning on the apoptosis of intrahepatic cholangiocyte and if ischemic preconditioning could decrease damage on intrahepatic bileducts for apoptosis through up-regulating expression of genes bag-1,bcl-2.Methods:30 SD rats were randomly divided into sham operation(SO)group,IR group,IP group,there were 10 rats in each group.Preparing IR and IP model according to Nauta.Left hepatic lobes tissue was sampled to determine bcl-2,bag-1 proteins and genes using immunohistochemistry and hybridization in situ.Liver samples were observed by pathematology.Apoptosis was discerned according to morphology in HE staining tissues to calculate AI.Datas were expressed by mean±standard deviation((?)±s),group comparison used t-test,multi-groups comparison used one-factor analysis of variance,there was statistic variance when P<0.05.Results:Cmpared with SO group,apoptosis increased in IR group.There was significant decrease of apoptosis in IP group compared with IR group(P<0.05).Expression of bag-1,bcl-2 mRNA and protein were both significane in intrahepatic cholangiocyte,IR group decreased and IP group increased.The result shows that IP may protect bile duct by elevating the expression of bcl-2 and bag-1.Conclusion:1.Bile duct injury initiated by IR in liver may relate to inducing apoptosis.2.At room temperature,IP may protect bile duct damage from IR.3.IP may protect bile duct from IR injury by elevating the expression of bcl-2 and bag-1 genes on large bile duct.
