Objective: We researched the protective effects and its Effects on Alteration of Protein Expression Pattern of Potentilla anserina L. alcohol extract following acute myocardial ischemia in RatsMethods: Male Sprague-Dawley rats were randomly divided into sham operation group and injury groups which were subjected to coronary occlusion lasting for 30min and then subdivided into model and diltiazem groups,intervention group of Potentilla anserina L. alcohol extrac(t7.2g/kg,3.6g/kg,1.8g/kg). The fuction study included analysis the ST segment change of Electrocardiogram after occlusion of LAD in Rats; record Influence of Potentilla anserina L. alcohol extract on homodynamic during ischemia injury in rat hearts. Observe the change of mitochondrion and glycogen in myocardial cell by electron microscope. Activities of LDH, CK, SOD and productions of MDA in serum were measured. The change of protein expression pattern in serum was monitored with weak cationic exchanger chips and strong anion exchange chips (SAX-2) by surface-enhanced laser desorption ionization-time of flight-mass spectrometry.Results: 1.intervention group of Potentilla anserina L. alcohol extrac(t7.2g/kg,3.6g/kg,1.8g/kg)could significantly Decrease the ST segment ascending; the diltiazem group also could decrease the ST segment ascdending. 2. The change of ultra structure in the myocardial cell: in the sham operation group the myocardial cell arrange in good order. The structure of mitochondrion is completed. The amount of glycogen in mitochondrion is abundant. The nucleuses are in good condition. In the model group, the myocardial cell arrange in disorder. There is severity edam at the edge of the nucleuses, the amount of glycogen in mitochondrion is significantly decrease, lots of mitochondrial crista are disappear or fusion. In intervention group of Potentilla anserina L. alcohol extrac(t7.2g/kg,3.6g/kg,1.8g/kg)and diltiazem group, the structure of the myocardial cell is better protected, there is plenty of glycogen in mitochondrion and there is less destruction mitochondrial crista. 3. 7.2g/kg dose and 3.6g/kg dose of Potentilla anserina L. alcohol extract could significantly degrade the activities of LDH (P<0.01), CK (P<0.05) and increase the activities of SOD (P<0.05) and inhibit MDA productions (P<0.01) in the serum of acute myocardial ischemia in Rats. But 1.8g/kg dose can not (P>0.05) compared with model group. Diltiazem group also could significantly degrade the activities of LDH (P<0.01), CK (P<0.05) and increase the activities of SOD (P<0.05) and inhibit MDA production (P<0.01) in the serum of acute myocardial ischemia in Rats. There is no significant difference between diltiazem group and high dose group. 4.Homodynamic: Myocardial functional parameters, left ventricular systolic pressure(Lvsp), maximum positive and negative change in pressure(士dp/dt) and left ventricular end diastolic pressure(Lvedp) were detected. Diltiazem group,7.2g/kg dose and 3.6g/kg dose of Potentilla anserina L. Alcohol extract could significantly improve Myocardial functional (P<0.05). There is no significant difference between 1.8g/kg dose of Potentilla anserina L. alcohol extract and model group of improving myocardial functional(P>0.05).5. Changes of protein expression patterns: (1) A total of 338 protein peaks were detected on WCX-2 chips and a total of 348 protein peaks were detected on SAX-2 chips in serum. (2) Among the total of protein peaks detected, the relative intensity of 27 protein peaks was shown to be differentially altered on WCX-2 chips in serum among model group,drug groups and sham group(P<0.05). The relative intensity of 16 protein peaks was shown to be differentially altered on SAX-2 chips in serum among model group,drug groups and sham group(P<0.05). There were 55 protein peaks in serum of drug groups and sham group which couldn't be detected in model operation group with WCX-2 chips. There were 47 protein peaks in serum of drug groups and sham group which couldn't be detected in model operation group with SAX-2 chips. (3) There were 55 protein peaks in serum of drug groups and sham group which couldn't be detected in model operation group with WCX-2 chips. There were 47protein peaks in serum of drug groups and sham group which couldn't be detected in model operation group with SAX-2 chips. (4)Compared with model operation group, the relative intensity of 4972Da protein peak increased following acute myocardial ischemia, the relative intensity of 4972Da protein peak decreased following acute myocardial ischemia in the intervention group of Potentilla anserina L. alcohol extract, which may be a target for the drug. (5)Compared with model operation group, there were 48protein peaks that showed the increase of protein expression and 44 protein peaks that show the reduction of protein expression on two kinds of chips in serum of drug groups and sham group.Conclusion: 1.Potentilla anserina L. alcohol extract could improve the myocardial function after acute myocardial ischemia in Rats. 2. The protein expression patterns of serum after acute myocardial ischemia in Rats were different, which sugge sted that part of protein in serum could be products of gene expression induced by acute myocardial ischemia. 3. Compared with model operation group, the relative intensity of 4972Da protein peak increased following acute myocardial ischemia in the model group, the relative intensity of 4972Da protein peak decreased in the intervention group of Potentilla anserina L. alcohol extract , which may be a target for the drug.
