Juema refers to the desiccated root of Potentilla anserine L.For medicinal use,it is neutral in nature and has the effect of supply vital force,invigorate spleen,beniefit lung,generate saliva and stop sputum.As a commonly used Tibetan and tonic medicine,Potentilla anserine L has always been used as a kidney tonifying medicine,moreover,it is only treated as ordinary food by local residents.The development and research of Potentilla anserine L and its polysaccharides are restricted by the location of habitat and cultural economy,so it is urgent to improve the deep processing and utilization level of fern polysaccharide.In this paper,the Potentilla anserina L.polysaccharide selenite-induced,which was isolated and purified in the early stage of the group,was optimized by single factor and response surface design,and the preparation factors were synthesized to synthesize fern polysaccharide selenite with different selenium content.The product was characterized by structure,and its antioxidant activity in vitro and anti-radiation function were studied.The above studies have not been reported in the same literature so far,the necessary experimental basis for further study of Potentilla anserina L.polysaccharide selenite-induced as an anti-radiation drug was laid.The results are as follows:?1?The optimum reaction time of Potentilla anserina L.polysaccharide selenite-induced is 134.84 min,temperature 78.39?,catalyst amount 49.37 mg,feeding ratio?Potentilla anserina L.polysaccharide:H2Se O3?=1:0.81.In this condition,the selenium content of Potentilla anserina L.polysaccharide selenite-induced prepared can reach 8518.81?g/g,The selenium content of the product was improved to a large extent.The most influential factors on selenium content are:catalyst amount>reaction time>temperature>feeding ratio.?2?Through analysis of UV,FT-IR and XPS,it is proved that five kinds of selenium fern content of linolenic acid ester structure contains Se=O bond and Se-O bond,that is,Potentilla anserina L.polysaccharide selenite-induced,the use of SEC-LLS and XPS characterization samples,The smaller the molecular weight,the lower and the thermal stability of the Potentilla anserina L.polysaccharide selenite-induced with higher selenium content were found.The monosaccharides composition and molar ratio of Se PAP1-5 were determined by GC-MS.It was found that each sample contained mannose and glucose.Se PAP1-3 contained a small amount of galactose,moreover,the mannose content increased and glucose content decreased with the increase of selenium content in each sample;The images of Potentilla anserina L.polysaccharide selenite-induced was studied in solution by atomic force microscope?AFM?.It was found that at low concentrations?0.01?g/m L?,branched polymeric chains were formed.When the concentration rose to0.05?g/m L,the polymeric chains were agglutinated to form globules and floccus,and when the concentration rose to 0.1?g/m L,they were bound into larger globules and irregular thick chains.?3?Using the method of phthalates self-oxidation to determine the ability of samples removal of·O2-,using salicylic acid method to determine the ability of samples removal of·OH,through the determination of sample removal DPPH free radical ability,it is found that fern polysaccharide Selenite has the ability to remove·O2-,·OH and DPPH free radicals,It was found that both Potentilla anserina L.polysaccharide and Potentilla anserina L.polysaccharide selenite-induced had scavenging ability for free radicals·O2-,·OH and DPPH,and the scavenging rate had a dose-dependent relationship with the sample concentration.The total scavenging capacity of Potentilla anserina L.polysaccharide selenite-induced was stronger than Potentilla anserina L.polysaccharide and was proportional to the selenium content.The results showed that selenization improved the oxidation activity of Potentilla anserina L.polysaccharide in vitro.?4?A 6 Gy dose of 60Co?-ray irradiation of RAW264.7 cells was selected to establish a cell model of radiation damage in vitro and to study the anti-radiation effect of Potentilla anserina L.polysaccharide selenite-induced.It was found that both Potentilla anserina L.polysaccharide and Potentilla anserina L.polysaccharide selenite-induced could enhance the SOD activity of RAW264.7 cells damaged by radiation,and the SOD activity increased with the increase of selenium content in the sample,and the MDA content of radiation damaged cells could be significantly reduced,and the MDA content increased with the increase of selenium content in the sample,and the GSH content of radiation damaged cells could be significantly increased,and the GSH reduced with the increase of selenium content in the sample.The results show that the Potentilla anserina L.polysaccharide selenite-induced can play a protective and repair role in the radiation damage of RAW264.7 cells caused by 60Co?-ray irradiation.On the whole,the anti-radiation effect of Potentilla anserina L.polysaccharide selenite-induced was better than that of Potentilla anserina L.polysaccharide with significant difference,indicating that selenization can enhance the anti-radiation activity of Potentilla anserina L.polysaccharide.
