Objective: Renshenqiangxindiwan(It was composed of renshen,shengdihuang,chansu medicine etc.)have efficacy of yiqiyangyin,huoxueqiangxin.It is a valid anagraph through my advisor clinical observation.It has fine therapeutic efficacy. To study the potential mechanism of renshenqiangxindiwan treating CHF, reference pertinent literature, to set up CHF Rats Model, at the same time,exert medicine intervention, to observe the effect on heart function and cardiac muscle cell apoptosis, to discuss mechanisms of renshenqiangxindiwan that to resist cardiac muscle cell apoptosis,to safeguard cardiac muscle cell and strengthen heart function. Moreover,we use HEdyeing to observe the effect on cardiac muscle histomorphology.Methods: Select Wister rats 50,weight from 160g to 180g,the female and the male were equal. Rats drink water and take food freely,and were raised in sanitary experiment lab that temperature was 18℃~22℃,bright and dim alternate every 12 hours. After 1 weeks,divided into 5 groups randomly:the normal group,the model group,the high-dose renshenqiangxindiwan,the low-dose renshenqiangxindiwan group,the xinbaowan control group. All the groups were made model and treated but the normal group. All the groups were lavaged drug which is calculated according to 100g·ml-1 one time each day. According to reference documents, the model rats were injected with ADR in abdominal cavity,which was dissolved into 1ml physiological saline according to 2mg·kg-1.One time a week, together six times. We injected ADR (2mg·kg-1) in abdominal cavity to the treating groups and equal dose of physiological saline to the normal group, The rats of xinbaowan and renshenqiangxindiwan groups were lavaged corresponding drugs for six weeks. The rats of normal group and the model group were lavaged physiological saline. At the end of the experiment, the rats were forbidden to eat for 12h. All rats were anesthetized by injecting 10% Chloral Hydrate into abdomen,tracheal intubation(tidal volume is 5ml)and artificial respiration(frequency is 10/min), draw off right common carotid artery and drive off breast and ascending aorta,all rats were injected 0.3% Calparine into vena caudalis,after heparinization,intubate right common carotid artery in order to record blood pressure,intubate apex of heart in order to record left ventricle pressure,to place number 2 electromagnetic flowmeter detecting head,to measure aortic blood flow through MF-27 electromagnetic flowmeter, and to record HR,ABP,ADP,SV,LVSP,±dp/dtmax,CO though RM-6000 eigh lead physiograph. We got tissue as size as mung bean from apex of heart, cleaning blood by physiological saline,bloting moisture content by filter paper, fixing by 70%spiritus vini, Put the tissue into refrigerator in order to test, We get immunofluorescence data analysis use Epics-XLⅡflow cytometer that produce from Beckman Coulter company of American and use Expo32ADC, and get fitting analysis of cell life cycle use MuticycleAV analysis-software. As the same time, to get the cardiac muscle tissue,HEdyeing,and to observe the change of pathomorphism of cardiac muscle under light microscope.Results:1 The common behavior of all the ratsDuring the process of making mode, the rats'color pattern of normal group was luster, drink and food normally, and having sensitive reflection.The model group rats were thin, thirsty, fur in disorder and withered,body to heaped up,activity to decrease,urine to decrease ,appetite to descend,oedema and onyx cyanosis etal sings and symptoms. After the treatment, the rats of treating group became well.2 Renshenqiangxindiwan's effect on pathomorphism of the cardiac muscleResult to show, the cardiac muscle cell of normal group was texture clear, shape was normal, endochylema's color was well-distributed, mesenchymal cell have no hyperplasy and no synathroisis. the model group rats'cardiac muscle fiber was not normal, part of the cardiac muscle to show apomorphosis, texture disappeared, mesenchymal cell was oedema in light degree, part of the cardiac muscle's shape was disorder. The cardiac muscle cellular nucleus was analosis, dyeing was darken. The treating groups'condition was more lighter than model group.3 Effect of renshenqiangxindiwan on heart functionThe heart systole and diastolic function of model group rat'was obstacled, the content of CO,LVSP,±dp/dtmax of model group rats'was decreased obviously than that of normal group (P<0.05or P<0.01).After the treatment,compared with model group,The content of CO,LVSP,±dp/dtmax of another group rats'was obviously rised(P<0.05or P<0.01).To show the heart function was improved.4 Effect of renshenqiangxindiwan on apoptosis rate compared with N group,the apoptosis rate of model group rats'was advanced obviously(P<0.05),After the treatment, compared with M group, the apoptosis rate of another group rats'was decreased obviously(P<0.01),compared with H group, the apoptosis rate of low dose group rats'was rised (P<0.05). There was no difference among high dose group and xinbaowan group (P>0.05).5 Effect of renshenqiangxindiwan on cardiac muscle cell generation cyclethe percentage of cell in G0/G1 stage of model group rats'was higher obviously than Normal group(P<0.05),but the percentage of cell in S, G2/M stage of model group rats'was lower than Normal group (P<0.05),After the treatment,the percentage of cell in G0/G1 stage of another group rats'was lower obviously than model group(P<0.01),but the percentage of cell in S, G2/M stage of another group rats'was higher than model group(P<0.05or P<0.01). The percentage of cell in G0/G1, G2/M stage of low dose group rats'was higher than high dose group(P<0.05),There was no difference among high dose group and xinbaowan group (P>0.05).compared with N group, the PI of model group rats'was lower obviously(P<0.05),After the treatment, the PI of another group rats'was higher than model group(P<0.01),it was show that the cell increment of treatment group rats'was higher than model group. compared with H group, the PI of low dose group rats'was lower obviously(P<0.05),the PI of xinbaowan group rats'was higher than low dose group (P<0.05),There was no difference among high dose group and xinbaowan group (P>0.05).6 Effect of renshenqiangxindiwan on Bcl-2 and Baxcompared with N group, The cardiac muscle cell apoptosis rate of model group rats'was higher obviously(P<0.01),but the FI of bax and Bax/Bcl-2 was rised(P<0.05), the FI of bcl-2 decreased obviously(P<0.05). After the treatment, the FI of bax and Bax/Bcl-2 of treatment group rats'was lower obviously(P<0.05), the FI of bcl-2 rised obviously (P<0.05), it was show that with apoptosis rate rised, The albumen express quant of bax rised,but The albumen express quant of bcl-2 decreased, the FI of bax and Bax/Bcl-2 of low dose group rats'was higher than high dose group(P<0.05),There was no difference among high dose group and xinbaowan group (P>0.05).Conclusion:1 Renshenqiangxindiwan could amendment affection of cardiac muscle histologic obviously, after treatment, result to show that the apomorphosis of cardiac muscle cell was abatement or disappearance.The cardiac muscle to line up in order, the mesenchymal cell have no synathroisis obviously. It proved that renshenqiangxindiwan could protect cardiac muscle cell.2 Renshen-qiangxin-diwan could improve every item of Haemodynamics,it is show that LVSP,±dp/dtmax,CO of treatment group rat'were better than model group, consequently,it could improve heart function of CHF rat'.3 Renshenqiangxindiwan could improve albumen express quant of bc1-2,and increase albumen express quant of bax,cell apoptosis rate decreased, cell apoptosis tendency to weaken,so it could protect cardiac muscle cell, to avoid the amount of cardiac muscle cell decreased,and to achieve affection of resist CHF.This is one of mechanism of action that renshenqiangxindiwan treat CHF.
