ObjectiveTo observe the effect of Shenqi Fuzheng Injection on Bcl-2 and Caspase-8 expression in cardiomyocyte apoptosis of rats with adriamycin induced congestive heart failure, which to provide evidence for clinic.Methods:Rat model of congestive heart failure was established by intravenous injection of Adriamycin according to its cardiac toxicity. Sixty male SD rats were divided randomly into 6 groups, normal group, adriamycin model group, captopril group, Shenqi injection(short by Shenqi following) low dose group, Shenqi middle dose group, Shenqi large dose group. The spirits, activities, eatings and color pattern were observed during the lab procedure. Eight weeks later, left ventricle heart indexs of rats were determined, and pathological changes in cardiac muscle by HE dyeing and expressions of protein Caspase-8,Bcl-2 in cardiac muscle of rats by immunity methods were observed. Caspase-8,Bcl-2mRNA in cardiac muscle of rats were semiquantitative analyzed by RT-PCR methods.Results:1. Myocardial hypertrophy index: Letventricrlarmass/bodyeight(LVM/BW) and myocardial hypertrophy index of left heart of rat model group were highed than the normal group(P<0.01). Each treatment group could improve myocardial hypertrophy in different degree than model group. The curative effect of Shenqi middle dose group was significant(P<0.01) and Shenqi large dose group was unknown significance(P>0.05)than model group. 2. Cardiac function Determination: the cardiac function of model group were lower than the normal group (P<0.01). Each treatment group could improve cardiac function in different degree than rat model group. The curative effect of Shenqi middle dose group was significant(P<0.01) and Shenqi large dose group was unknown significance(P>0.05).3. The expression of protein Caspase-8,Bcl-2: the results of immunity methods showed that the expression of protein Caspase-8,Bcl-2 in group model group were all obviously higher than normal group(P<0.01). Each treatment group could decrease the expression of apoptosis protein than the model group.The expression of Shenqi middle dosage group were significantly decreased(P<0.01)and Shenqi large dosage group were unknown significant difference(P>0.05).4. The results of RT-PCR:Compared with normal group, Caspase-8mRNA of model group were significant increased(P<0.01)and Bcl-2mRNA were significant decreased(P<0.01).The expression of Caspase-8mRNA in small and middle dosage group and captopril group were significant lowed than the model group(P<0.01, P<0.05).The expression of Bcl-2mRNA in above group were significant highed than the model group(P<0.01, P<0.05). The expression of Caspase-8mRNA,Bcl-2mRNA in large dosage showed no significant difference(P>0.05).Conclusion:1. It is effect that rat model of congestive heart failure was established by intravenous injection of Adriamycin according to its cardiac toxicity.2. Shenqi injection can improve the cardiac function of rat with congestive heart failure.3. Shenqi injection can lessen the cardiomyocyte apoptosis in rats with congestive heart failure.4. Shenqi injection can improve the expression of protein Caspase-8,Bcl-2 in the cardiac muscle of rat with congestive heart failure. 5. Shenqi injection can decreased the expression of Caspase-8 or inhibit its activity, and increased the expression of Bcl-2 in the cardiac muscle of rat with congestive heart failure.6,Analisied Compared with Caspase-8/Bcl-2 and cardiomyocyte apoptosis teneise realet。...
