Objective: To study the expression of Livin in bladder transitional cell carcinoma(BTCC) and the correlation between expression of Livin and clinicopatholgicalparameters (pathological grade, clinical stage and recurrence); To research the relationof Livin, PCNA and P-GP; To disclose the molecular mechanism and drug resistencemechanism of BTCC in order to provide new clinical evidence for the prognosis andtherapy of BTCC.Methods: Immunohistochemistry (IHC) (SP) was used to detect the expression ofLivin, PCNA, P-GP in BTCC(n=48) and normal bladder specimen (n=10) and thenfind the the correlation between their expression and clinicopathological parameters.Results: IHC indicates that integrated optical density (IOD) of livin in BTCC is6.81±2.34(×10~4), while the control is 0.92±0.54(×10~4). There are remarkabledifference between them, there are no visible relation between Livin and tumor'spathological grade and clinical stage (P>0.05). But Livin correlates with therecurrence of BTCC. PCNA index (PI)(%)in BTCC is 55.81±22.95, while the controlis 13.40±9.62, there are remarkable difference between them (P<0.01). PCNA deeplycorrelate with tumor's pathological grade (P<0.05) and recurrence (P<0.01) buthas no difference from clinical stage (P>0.05). IOD of P-GP is 5.13±2.34, while thecontrol is 1.25±0.86, the difference is remarkable (P<0.01), P-GP in BTCC correlateswith clinical stage and recurrence (P<0.01), but has no relation with tumor'spathological grade (P>0.05). These research indicates that Livin positively correlateswith P-GP and PCNA(r=0.64, r=0.76)Conclusions: The expression of Livin in BTCC doesn't correlate with the grade andstage but deeply correlates with the recurrence, indicates that the expression of Livin may become one of the indexes which can be applied to evaluate the recurrence risk,and may supply theoretical basis for gene targeted therapy after operation; PCNAdeeply correlates with BTCC's pathological grade and recurrence, and positivelycorrelates with Livin, indicates that the tumor's develop and recurrence may be theoutcome of coordinate of proliferation and apoptosis, may be they have commonpassway in the recurrence of BTCC. When united we may improve the accuracy ofevaluating the recurrence risk; The expression of P-GP in BTCC correlates withclinical stage and recurrence, and positively correlates with Livin, indicates that theymay make effects in drug resistence through multi mechanisms and may havecommon passway, we should treat the drug resistence by multi therapies. When putLivin, PCNA, P-GP together we find that they are more credible than any one or twoof them united when applied to evaluate the recurrence risk, so it may be a new indexfor evaluating the recurrence after operation.
