Objective: Apoptosis, which could maintain homeostasis and lead to orderly celldeath process with certain genetic control. And the family of IAPs which can inhibit cellapoptosis through the endogenous and exogenous way, especially in cancer cells with aparticularly obvious expression, have a close relationship with cell proliferation andapoptosis,and play a very important role in antiapoptotic of cancers.At present research,there’re all about8human IAPs found, including NAIP, cIAP1/2, XIAP, Survivin,BRUCE, ILP2and Livin. Bladder cancer is the most common malignant in urinarytumors,especially transitional cell carcinoma, including superficial bladder cancer andcurrent invasive bladder cancer in clinical. And invasive bladder cancer has a highdegree malignancy, easy to transfer, and has a poor prognosis. The present study hasproved that Livin, Survivin, XIAP, cIAP1/2have all expressed in the bladdertransitional cell carcinoma (BTCC). Researches show that Livin, XIAP and cIAP2express in the cytoplasm, Survivin and cIAP1express in nucleus. That Survivin andcIAP1are not only association with the apoptosis of cancer cell, but also have a relationto cell cycle. Thus, to identify the expression of IAPs in BTCC, will provide anextremely important suggestion on the diagnosis, therapy and evaluation of prognosis ofBTCC. However, the regulation of IAPs on the proliferation and apoptosis, the internalcorrelation among multi-IAPs while expression in bladder carcinoma simultaneously,whether IAPs,and which could be the prognosis index, are unknown.Some researchesshow that the apoptosis of tumor cell is an extremely complex process, the apoptosis ofthe tumor cell is inhibited by IAPs, which can block the caspase pathway. Nevertheless,the biological fuction of inhibition by IAPs is not only the fuction of one IAP, it shouldbe the fuctions by multi-IAPs, they executive the process of apoptosis by"co-expression". Consequently, in the past research foundation about single IAP, weresearch the conditon of co-expression of Livin, Survivin, XIAP and cIAP1/2in thesamples of bladder carcinoma by immunohistochemistry and immunofluorescence, additionally, we also analyse the internal correlation among IAPs.Methods:1.Collecting specimens of BTCC from our hospital, then samples of paraffin,section, immunostain. Searching the conditon of co-expression of Livin, Survivin,XIAP, cIAP1/2in samples of BTCC.2.Human Cell line T24of BTCC culture, determine locations and conditions ofLivin, Survivin, XIAP, cIAP1/2by immunofluorescence.Results:Immunohistochemistry1.Livin, XIAP, cIAP2expressed in cytoplasm of BTCC cell. Survivin and cIAP1expressed in both nucleus and cytoplasm.2.There’re significantly associations between co-expressions of IAPs and clinicalstage and pathology classification.3.In BTCC, XIAP has a positive correlation with cIAP1-C, and Survivin-N has apositive correlation with cIAP1-N, by Spearman’rho.ImmunofluorescenceLivin, XIAP, cIAP2expressed in cytoplasm of cell line T24. Survivin and cIAP1expressed in both nucleus and cytoplasm,mainly in nucleus.Conclusion:1.There’re significantly associations between co-expressions of IAPs and clinicalstage and pathology classification.2.The evaluation of IAPs could be indexes for estimating the progression andfeature of clinical and pathology of BTCC or other malignant tumors.3.In BTCC, there’re some correlations among IAPs,which could regulate apoptosisand proliferation of tumor cell.
