Abstract/Summary:
AIM: To observe the pathological features, expression of tumor necrosis factor-alpha(TNF-α) andα-Smooth muscle actin (α-SMA) in liver tissue with cholestatic liverfibrosis in rat.METHODS: Cholestatic liver fibrosis model was induced by common bile ductligation(BDL) in rats. The contents of hydroxyproline(Hyp), hyaluronic acid(HA) andlaminin(LN) in liver tissue were determined 4wk after BDL. Meanwhile, thepathological changes of the liver tissues were observed under light microscope. Theexpression of TNF-αandα-smooth muscle actin(α-SMA) were detected byimmunohistochemical LAB-SA methods.RESULTS: Compared with that in the sham group, the contents of Hyp,HA and LNwere significantly elevated (P<0.001)4wk after in BDL group. Most hepatic lobularareas had been replaced by proliferated bile ducts, fibrous tissue, and biliary cirrhosiswas observed, theα-SMA positive cells aggregated prominently in the peripheral ofbile ducts, fibrotic tissue and septa, and the TNF-αpositive cells were prominentlyexpression in hepatocytes in BDL group.CONCLUTION Biliary cirrhosis developed after 4 wk of BDL in rats, and theproliferating bile ducts and fibrous tissue were a features. Theα-SMA positive cellswas coincident with collagen deposition in BDL group. The distribution of TNF-αandα-SMA positive cells were different in liver tissue, and its showed that TNF-αmay be associated with indirect activation of HSCs in rats biliary cirrhosis...
