Experimental Research On Effect Of Tiaogan Granule On The Gene Expression Of Tumor Necrosis Factor-α And Leptinmrna In Non-Alcoholic Fatty Liver Rats

Objectives: The Experimental research is designed to observe the influence of the the Tiaogan Granule on the serum of TNF-α,LP content and its liver tissue gene expression levels of the rats with nonalcoholic fatty liver disease, and study the mechanisms. It will provide the scientific basis for that the Tiaogan Granule's becoming a new Chinese patent drug.Methods: Seventy-one of the eighty Wistar rats were randomly selected for modeling. After fed by high-fat diet for ten weeks, eight rats were randomly selected to take pathology to determine the success of the modeling. After the success of the modeling, sixty-three rats were randomly divided into such seven groups, with nine rats every group, as the model control group, diet control group, Kanra control group, seven March fatty liver pills control group, adjusted liver granules low, and medium high-dose group, and the rest belong to the normal control group. Then came the observation of the general state of rats before and after the treatment and liver function, blood lipid, MDA, SOD; the kidney histopathologywas HE stained animal model of pathological change after the treatment; the two-step method of immunohistochemistry was adopted to test portein expression of liver tissues CYP2E1, TNF-α, using real time - polymerase chain reaction (Real Time-PCR) assay of liver tissue TNF-α, and LP mRNA expression.Results: 1. Tiaogan granules can reduce body weight and liver weight index;2. Reduction of serum ALT, AST, TC and TG levels; 3. Regulate liver tissue MDA, SOD activity; 4. Reduce the liver tissue serum of TNF-α,CYP2E1And express the amount of protein;5 .Reduce the liver tissue of TNF-α, LP or its gene expression levels, To alleviate the pathological changes in liver tissue tended to be normal.Conclusion: The experimental results show that Tiaogan Granule has obvious effect on anti-lipid peroxidation, decreasing body weight, liver weight index and liver inflammation, and lowering the level of the serum ALT, AST, TC and TG. Therefore, the pathological changes in liver tissue gradually returns to normal condition. The mechanism of Tiaogan Granule to produce these effects is closely related to its function of regulating the level of tumor necrosis factor-αand leptin. All above, Tiaogan Granule is highly effective against the nonalcoholic fatty liver disease, and thus is worthy of further research.
