BackgroundIt is many years when D. Huang and J. G. Fujimoto et al used the OCT to imagethe eye in 1991. They found that the OCT can image the microstructure of the eye.And from then, there are many people researching OCT. For example, some peopleresearch its system (include how to improve the axial resolution,image-velocity,image-depth of this system and so on); Some groups research that other parameterschange when the light go through the tissue by OCT (such as: Doppler OCT, PS-OCT,Spectrum OCT, Differential absorption OCT et al.); There are also some other peoplewho mainly study on the application of the OCT, researching the OCT apply indifferent tissue, and distinguish between normal tissue and abnormal tissue. Thus theOCT can help the doctor diagnosis.Although the research of OCT has given great advancement, to now, there aresome questions of OCT researching. For example: some people only research whetherthe OCT can image some tissues, and make the images of the OCT compare thepathology slices; or researching the things which can improve the depth of the OCTimaging. But there are few people who use the OCT to real-time image the processesof the disease. And now the hospital is lack of the system which is a non-invasive,high-axial resolution, non-contact and real-time. Thus this paper mainly resolves thequestions.PurposeIn this thesis, we mainly investigated that using OCT to image the rat's tongue,the mode of the skin ulcer and the mode of the acetic acid gastric ulcer to probe intothe application worth. In order to provide a base for making OCT apply in theinspection of tongue of the Traditional Chinese Medicine, and all kinds of the ulcerdiseases.Materials and methods The OCT system was used in this study, in which a broadband light source withthe central wavelength at 1310 nm and a bandwidth of 50 nm is used at wavelengths inthe therapeutic window. The light source yields a 151am axial resolution in free spaceor approximately 10μm in the tissue if the mean refractive index of bulk tissue isassumed to be 1.38. The signal-to-noise ratio (SNR) of the system was measured at100 dB. And using the Lab View soft controls the operation. The main experiments ofthis thesis are following by OCT:1) Using OCT examine the tongue of the rat: In the present study, in order to maketongue of the Traditional Chinese Medicine become a objective and accuratediagnostic method, we use an OCT system to examine the tongue of the normalrat. After examination, the tongue was surgically removed, sectioned by amicrotome and put on a glass slide for histological examination.2) Using OCT to monitor the healing processes of the skin wound and skin scald: Inthis experiment, we create the modes of skin wounds and skin scalds in the rats.Let the wound heal spontaneously. Then using the OCT monitor the healingprocesses of skin wounds and skin scalds in the rats on the next day and everytwo days, thereafter to measure the depth and degree of wound healing.3) Using OCT to monitor the model of acetic acid gastric ulcer in rats: Making themodel of acetic acid gastric ulcer in rats by acetic acid, and then the healingprocess of acetic acid gastric ulcer in rats was observed by OCT on the secondweek, and every one week image rats by OCT. The ulcerative gastric mucosatissues were detected by OCT, and then the imaging on ulcerative gastric mucosatissues were analyzed. The experiment continues about 4 weeks.Results1) The result of using OCT examine the tongue of the rat:Results of the OCT imaging studies showed that the OCT system was capable ofshowing the images of three distinct regions of the tongue: the tongue coatinglayers, the interface between the tongue coating and the tongue body, and thetongue body. It was also possible to assign an index number to each of the coating layers. When the tongue tissue was examined under a microscope, three separateregions of the tongue were also visible. Thus tongue can become objective.2) The result of using OCT to monitor the healing processes of the skin wound andskin scald:The healing stages of the skin wounds and the skin scalds could be clearlyobserved from the OCT images. Wound healing of rat skin may be arbitrarilydivided into the following stages: (a) wound generation, (b) suppurating in someinflammation area, (c) growing of granulation tissue, (d) scab formation, (e)decrustation, and (f) completely healing. The wound depth and the extent ofhealing also could be determined from the OCT images in the different healingtime. Healing time for the skin scald was found to be much shorter than those ofthe skin wound. Because the skin wound is serious than the skin scalds.3) The result of using OCT to monitor the model of acetic acid gastric ulcer in rats:Results of this experiment showed that using OCT technology can identify theulcerative gastric mucosa tissues with normal tissues, make the imaging on theunder-layer of gastric mucosa tissues, and evaluate the healing period and healingquality. From the OCT images can be clearly found the acetic acid gastric ulcer. Inthe last stage, the changes of the under-layer of gastric mucosa tissues are clearlyobserved.Discussions1) Based on the results of this preliminary study (using OCT examine the tongueof the rat), the OCT can distinguish the microstructure of the rats tongues.Thus the OCT potentially can be developed into a glossoscopy for clinicalapplication in TCM practice.2) Results of this study (using OCT to monitor the model of acetic acid gastriculcer in rats) demonstrated that OCT technology is a promising scientific toolin helping to make diagnostic decisions and recommend treatment strategy tbrskin wound and skin scald since the technology can provide a real-time, high axial resolution, and fast imaging on wound depth and the degree of healing.The OCT can be used estimate whether all kinds of the skin ulcer diseases arecompletely heal up.3) Based on the results of this preliminary study (using OCT to monitor themodel of acetic acid gastric ulcer in rats). We can get this good result, and ifusing endoscopic-OCT can help doctors make diagnostic decisions on pepticulcer, and evaluate the healing quality and the probability of gastric ulcerrecrudescing.
