Objective: To establish animal model for echinococcus granulosus infection,and to obvserve the hydatid cyst with HE and to investigate the expression and distribution of osteopontin (OPN) in hydatid cyst.Methods: Firstly a half of mice were injected hydrocortisone as to make a non-specificity immunizing inhibitive animal mddel.Secondly E. g suspension was injected through three paths to establish E. g model as follows(1)portal vein inoculation,(2)abdominal opening injection,(3)percutaneous transhepatic inoculation.At last,the hydatid cyst was observered with HE and immunohistochemistry staining by using antibodies specific for OPN.Results: E.g infection rates of normal immunizing mice were 93.1% in group (1),80.2% in group (2) and 59.3% in group (3).The infection rates of non-specificity immunizing inhibitive mice were similar with normal immunizing mice. It's obviously disparity between hydatid cyst and macrophages in HE. OPN was selectively distributed in macrophages while it was approximately not found in hydatid the cyst.Conclusion: E.g animal model can be established by above three methods.Portal vein inoculation has higher infection rate and more encysts than that of other two methods. Animal model shows that there is a partible interspace between hydatid cyst and liver parenchyma. OPN plays a important role in hydatid cyst's formation mechanism.
