Effects Of Different Therapeutic Methods And Its Typical Recipes On The Expression Of CD14mRNA Of Liver Kupffer Cell In Rats With Fatty Liver

Objective: To observe the effects of different TCM therapies: soothing liver,invigorating spleen, dispelling dampness, promoting blood flow and complex recipeson serum lipid, liver function, liver histopathology and the expression of CD14mRNAof liver Kupffer cell in rats with fatty liver.Methods: The fatty liver model of rats was established by feeding withhigh fat diet and alcohol. Meanwhile, rats were given recipes of soothingliver, invigorating spleen, dispelling dampness, promoting blood flow andcomplex recipes respectively. 12 weeks later, to observe the influence ofdifferent therapeutic ways and corresponding recipes on serum lipid,liver function,pathology of liver,the expression of CD14mRNA on liver KCs in rats with fattyliverResults:1. Compared with uncontrolled group, the contents of serum TC,TG,ALT andAST in controlled group significantly increased(P＜0.05). After the intervention ofdecoction, the contents of serum TC,TG,ALT and AST in each treatment group werelower than those in controlled group, and those in soothing liver and invigoratingspleen group were the lowest (P＜0.05).2. In controlled group, light microscope observation revealed fatty degenerationfrom medium to severe degrees in hepatocytes: swelling of hepatocytes and differentsizes of lipid droplets inside the cytoplasm. Infiltration of inflammatory cells andmuch focal proliferation of fibers were also shown in the portal area of controlled rats.Compared with the controlled group, the degrees of fatty degeneration in treatmentgroups were significantly ameliorated, especially in the invigorating spleengroup. Swelling of hepatocytes, infiltration of inflammatory cells and proliferation of fibers in treatment groups were also ameliorated in different degrees.3 The expression of CD14mRNA on liver KCs: The level of controlled groupwere significantly higher than the one of uncontrolled group (P＜0.05);compared with controlled group, The levels of expression are all decreasedin invigorating spleen, dispelling dampness, promoting blood flow andcomplex recipes (P＜0.05), but there was no significant difference insoothing liver group; Among all treatment group, the expression in theinvigorating spleen group were the weakest (P＜0.05), but among dispellingdampness, promoting blood flow, complex groups, there was no significantdifferences.Conclusions:1. Eating too much high fat diet or drinking too much alcohol were the mainpathogenic factors of the fatty liver. Disturbance of the spleen function and phlegm,wet,stasis congealing were the main pathogenesis of the fatty liver. phlegm,wetand stasis were factors of the fatty liver2. Therapies of invigorating spleen are the main therapeutic methods of the fatty liver,and the therapies of dispelling dampness,promoting blood flow played veryimportant roles in preventing and treating the fatty liver.3 Therapeutic function of different TCM therapies to fatty liver were:3.1 Adjusting lipometabolism: lowering the level of serum TG,TC.3.2 Decreasing fatty degeneration of liver cell3.3 Improving liver function: lowering the level of serum ALT,AST.4 Restraining Kupffer cell function of fatty liver rat: invigorating spleen, dispellingdampness, promoting blood flow and complex recipes could down-regulate theexpression of CD14 of liver KCs in rats with fatty liver, restrain the activation ofKupffer cell, which maybe the main mechanism for preventing and curing fatty liver.
