Objective: To observe the effects of different therapeutic methods and the recipes of Traditional Chinese Medicine(TCM): soothing liver, invigorating spleen, promoting blood circulation, dispelling dampness and complex recipes on the activation of c-Jun N-terminal kinase(JNK) protein of Kupffer cells of fatty liver rats.Methods: The fatty liver rat models were reproduced by feeding with high fat diet combinated with distillate spirit via gavage, meanwhile the rats were administered medicinal solution of soothing liver, invigorating spleen, dispelling dampness, promoting blood circulation and complex recipes respectively for 12 weeks, and then the serum TC, TG, LDL-C, ALT, AST, etc. were analyzed, the histopathological changes of hepatic tissue were observed and the expression of JNK protein and phospho-JNK protein was detected by Western blotting.Results:1. Compared with normal group, the contents of serum TC, ALT and AST in model group were significantly increased(P<0.05). Compared with model group, the contents of serum TC, ALT and AST in all treatment group were decreased, significantly in soothing liver group and invigorating spleen group(P<0.05).2. Pathology of hepatic tissue: The fatty degeneration of hepatic tissue of the model group was from medium to severe degree, and there was significant difference compared with normal group(P<0.05), which indicated that the reproduction of fatty liver models was successful. Compared with model group, the degrees of fatty degeneration in soothing liver group and invigorating spleen group were significantly ameliorated(P<0.05), which indicated that the therapeutic methods of soothing liver and invigorating spleen had the effect of preventing and treating fatty liver. However, compared with model group, differences of fatty degeneration among dispelling dampness group, promoting blood circulation group and complex group had no statistic significance(P>0.05).3. The expression of JNK protein and phospho-JNK protein in Kupffer cells of model group were higher than that of normal group. The expressions of JNK protein and phospho-JNK protein of the treated groups were reduced in different levels; the expresstion of JNK protein was significantly depressed in invigorating spleen group, and the expresstion of phospho-JNK protein was significantly depressed in soothing liver group.Conclusions: 1. Improper diet is the main pathogenic factor of fatty liver. The functional disorders of liver and spleen in TCM theory were the fundamental pathogenesis.2. The therapeutic methods of soothing liver and invigorating spleen are the fundamental therapeutic methods of fatty liver.3. The results indicated that the above therapeutic methods and recipes, especially the therapies of soothing liver and invigorating spleen, might have preventive and treatment effects on fatty liver, such as adjusting lipid metabolism, ameliorating the fatty degeneration of hepatocytes, protecting hepatocytes, ameliorating inflammation.4. The high expression of JNK and phospho-JNK of Kupffer cells involve in fatty liver and contribute to liver impairment. The recipes of invigorating spleen and soothing liver may protect against liver injury by reducing the total JNK protein and activation of JNK protein of Kupffer cells in fatty liver model rats. And the mechanisms need further research.
