Objective In order to supply experimental and theoretical guide for improving the diagnosis,treatment and predicating the prognosis of Cervical Carcinoma , the expression and their relationships of NF-κBp65 and uPA protein in Cervical Carcinoma were investigated.Methods The expression of NF-κBp65 and uPA protein in 60 cases of cervical cell carcinoma ,20 case of cervical intracpithelial neoplasia and 30 cases of cervicitis were detected by immunohistochemical technique and its relation with tumor clinicopathological characteristics was analyzed.Results1. The expression levels of NF-κB and uPA protein in Cervical Carcinoma were higher than those in cervicitis(both P< 0.01).2. The overexpression of NF-κB activation was correlated with tumor invasion-related lymph node metastasis and the extent of tumor invasion(P<0.01),but NF-κB activation was not correlated with high pathological grade(P>0.05),clinical stage(P>0.05) and age(p>0.05).3. The overexpression of uPA protein was correlated with clinico-pathological characteristics of Cervical Carcinoma such as lymph node metastasis(P<0.01),clinical stage(P<0.05),pathological grade(P<0.05), the extent of tumor invasion. But the overexpression of uPA protein was not correlated with age.4. There was significant correlation between the expression of NF-κB and uPA in Cervical Carcinoma. The expression levels of NF-κB in Cervical Carcinoma were high ,so were the expressiong levels of uPA. The expressiong levels of uPA in Cervical Carcinoma were high , while the expression levels of NF-κB were low. Conclusion The expression levels of NF-κB and uPA protein in Cervical Carcinoma were higher than those in cervicitis.The overexpression of NF-κB activation was correlated with tumor invasion-related lymph node metastasis and the extent of tumor invasion (P<0.01), but NF-κB activation was not correlated with high pathological grade (P>0.05),clinical stage(P>0.05) and age(p>0.05). Constitutive activation of NF-κBp65 and the overexpression of uPA were correlated with biological behaviors of Cervical Carcinoma and suggested that NF-κB activity was related to tumor progression due to its transcriptional regulation of uPA and NF-κB may be a key factor for the happen and development in Cervical Carcinoma,which plays an important theoretical significance in the treatment and predicating of Cervical Carcinoma.
