Objective To study the effects of acute hypervolemic hemodilution with 6% Hydroxyethyl Starch at different doses on the expression of tumor necrosis factor(TNF-α) and E-selectin after renal ischemia-reperfusion in rats.Methods Forty male SD rats were randomly divided into four group with ten rats in each group:group ischemia-reperfusion, in group 2-4 was infused 7.5ml·kg-1, 15ml·kg-1, 30ml·kg-1 before ischemia-reperfusion. One femoral vein was cannulated for continuous intravenous infusion for thirty minutes. After infusion, each group was subjected to 30 min ischemia and followed by 4h reperfusion. TNF-αlevel in the blood plasma were measured with enzyme-linked immunosorbent assay(ELISA)system and the expression of E-selectin on rat renal tissue were detected by immunohistochemistry staining.Results HES 7.5ml·kg-1, 15ml·kg-1 significantly reduced the TNF- a level in the blood plasma. The level were 25.76±7.59 and 34.15±8.83. (P<0.05).HES 7.5ml·kg-1 reduced the TNF-αlevel in the blood plasma comparing with HES 15ml·kg-1 (P<0.05). The expression of E-selectin in the tissue significantly reduced to 152.78±2.89 in HES 7.5ml·kg-1 ,154.91±3.49 in 15ml·kg-1, comparing with group ischemia-reperfusion(P<0.05).Conclusion HES can reduced the injury of ischemia-reperfusion, this reduction was due to the dose.
