To explore how voluven influences cardiovascular function and its mechanism in myocardial ischemia-reperfusion(I/R) rats.Rat's I/R model was established by ligating left anterior descending coronary artery(LAD) for 30 min followed by reperfusion for 180 min. To investigate protective effects of voluven in I/R rats by evaluating hemodynamics at different time points and observing histopathology of each group. The serum concentration of TNF-α, IL-1βand the change of activity of nuclear factorκappa B(NF-κB) were determined through ELISA; ICAM-1mRNA in rat heart was quantified through real time PCR ,and the morphologic features of apoptosis were identified histochemically by TdT-mediated dUTP niked labeling (TUNEL) staining,through which we studied molecule mechanisms of protective effects of voluven on myocardial I/R rats.The results show that voluven could improve cardiovascular function and attenuate I/R injury, and the mechanism may be that voluven inhibits the activity of NF-κB in I/R heart and decreases infiltration of PMN.
