The veterbral body fracture is the ealiest,most familiar complication of osteoporosis, under general therapy, it is not only ineffective but also produceing secondary fractures easly. As members of the TGF-βsuperfamily of factors Bone morphogenetic proteins (BMPs) are multifunctional growth factors that mediate a variety of biological functions, including bone formation. At present, the mechanism is dimness that BMPs are accelerating the osteoporotic veterbral fracture healing. Published data regarding the action of BMP often relies on studies using recombinant human BMP(rhBMP). However, the costs is difficult to sustain and The application is difficult to generalize in clinically. We adopted enough,inexpensive bovine BMP of yunger yak in Qinghai to accelerate bone defect and nonunion healing. We designed an animal experiment in an effort to determine if there were correlation between in roentgenography,histology,mechanics and the expression of VEGF ,to study the mechanism of accelrateing to heal of osteoporotic vertebral fracture and the feasibility of generalizeing in clinically.Aim: To study the biological function of vascular endothelial growth factor(VEGF),the mechanism of accelrateing to heal of osteoporotic vertebral fracture and the feasibility of generalizeing in clinically.METHODS①The preparation of bovine bone morphogenetic protein: The femur of yunger yak in Qinghai were crushed under -4℃with the method of Urist.②129 8-month-old female Sprague-Dawley(SD) rats were randomized into A,B,C three groups , A,B groups were 51 each, C group was 27.③A group was placed with the bovine bone morphogenetic protein group and B,C group was the nothingness model after 3 months.④The three rats were put to death after 3, 7, 10, 14, 21, 28, 56, 84 d, the callus of each rat was examined immunnohistochemistry to detect the expression of VEGF , also using image analysis to deal with it. After 1, 2, 4, 6, 8, 12weeks, it was analized with radiograph,histology and mechanics in 6, 8, 12weeks. All values were expressed as means±SD. One-way analysis of variance was used to assessgroup means, Pearson method to correlation analysis. P<0.05 was considered statistically significant. Statistics were using SPSS10.0.RESULT①Iimmunnohistochemistry: VEGF could be seen positive 7 days after doing bovine bone morphogenetic protein, and there had the highest quantities about 7-14 days post-transplantation. However there were not obviously high quantities in B group.②Radiomorphometric outcomes: The lumbar 5(L5) was pervious to light postoperatively. the area of the veterbral fracture of A,C groups were different from B group in 6 weeks postoperatively but same as in 8 weeks.③Histology: The chondrocyte were seen in one week. In A,C groups, fibrous callus postoperatively were formed in 7d ,regenerate bone and bone were merged in 21d, but in 56—84d, cartilaginous callus was replaced by osseous callus, marrow was repassed in fracture part, and the bone was mature; But in B group, we didn't fined soul fibrous callus postoperatively at 7d , regenerate bone were more isolate or dissociate and reticulate structure were existed at 28d, the defect bone weren't healing in 56~84d, the fracture areas were put off healing; (4)The results in compression testing of vertebral column were significant differences between B and A,C groups (p < 0.05) , but between A and C was similar. (P > 0.05) .CONCLUSION: (1)The skeleton is loosen in osteoporotic veterbrea, the cancellous were sparsity,lamellar,isolate or dissociate, the filling was many adipose cells, testifies in decrease of mechanical strength,quality of fracture healing and fracture easly.(2)BMP +FS can be used effectively to enhance vertebral fractures healing with respect to mechanical and histology bone properties, testifies in increase of mechanical strength or quality of fracture healing and in decrease of fragile bone. It was similar to C group.(3)BMP+FS can be used effectively to accelerate The chondrocyte to bone cell, enhance the trabecular to formation and the numbers, tightness, adipose cells were very small. regenerate bone were more less isolate or dissociate and reticulate structure, accelerated regenerate ossification.(4)It was positive that BMP +FS can be used effectively to enhance the compound and secretion of VEGF and accelerate fracture healing. VEGF play very important roles in accelerating fracture healing of osteoporosis rats.(5)Regenerated ossification of bBMP accelerated wasn't perfect mechanic in ealy but same as commmen veterbrea postoperatively, even the cherectures of fabric and physiology, it wasn't obvious humful that bBMP produced excluding reaction of immunization and disease of infectivity or epidemic. so bBMP was feasibility of that popularizeing and applying to after evaluation in clinically.
