Background and ObjectiveRecently,the attack rate of hyperuricemia and gout increasingly upgrade,it has become a common disease.To avoid the formation of gouty arthritis and renal lesions, patients have to take medicines of degrade uric acid in long-term, at present still short of safe and effective medicine in dealing this disease. Western medicine mainly use Non-steroidal anti-inflammatory drugs, colchicine allopurinol,this kind of medicine can relieve symptom, but have side effects,including gastrointestinal tract effect,and ruin nephridial tissue then affect it's function, cohmonly patient cann't tolerance. Chinese herbal medicine has long history in treating gout,but up to now there have no Chinese drugs pharmaceutics which can degrade the blood uric acid effectively in clinical.So,to development drugs that can degrade the blood uric acid has significant meaning, especially resort to chinese medical science experience.The purpose this experiment is to create a Hyperuricemia model that is similar to human physiological function,then base on this model test therapeutical effect of prescription HERBA SIEGESBECKIAE capsule in treating Hyperuricemia and uratic renal injury,which is professor yu-xuefeng's clinical empirical formula.1. To create a Hyperuricemia modelUp to now, there is no standard in create a Hyperuricemia animal model.It is generate excessive or hypoeccrisis about uric acid that induce Hyperuricemia. because of mankind have no uricolytic ferment,they cann't decompose uric acid,so in human being's it mainly for uric acid hypoeccrisis that lead to this disease, forthcoming models cann't eliminate the difference between human being and animal,this experiment is to create a Hyperuricemia animal model that is similar to human physiological function using adenine and Oxonic Acid.Using different density adenine and Oxonic Acid intragastric rat,detecting the content of serum uric acid, creatinine, urea nitrogen before and after experiment,to observe nephridial tissue texture's alteration.(1) High,middle and low dosage group's blood uric acid step up differently,high dose group's renal injury gravely, low dosage group' have no renal injury.(2) Middle dosage group's nephridial tissue texture's alteration: proximal convoluted tubule microvilli reduce obviously, cellula epithelialis's karyopyknosis lightly,cytolysosome accrementition, mitochondria membrane deficiency, spine dissolve or break.High dose group's proximal convoluted tubule microvilli disappear,part cellula epithelialis karyopyknosis,intracytoplasm cell organ decrease,cell organ's structure cloudiness,urinary cast intracavitary have bulky urate crystal. Low dosage group's renal tubular epithelial cell cytolysosome accrementition slightly, between canoula fibroblast fibroblast fibroblast.middle dosage group is suitable for the model of Hyperuricemia.2. The protective effect of prescription HERBA SIEGESBECKIAE capsule on Hyperuricemia animal modelDifferent dosage traditional Chinese medicine(low,middle, high) and allopurinol with making model drugs supply to every group respectively,detect the content of serum uric acid,creatinine, urea nitrogen before and after experiment, observe nephridial tissue texture's alteration.(1) Traditional Chinese medicine(middle, high) and Western medicine all can restrain the content of serum uric acid,creatinine, urea nitrogen;lower dosage traditional Chinese medicine has the function of postpone it's step up,has significant deviation in compare-with control group((P<0.01).(2) Middle dosage group of traditional Chinese medicine's kidney structure is normal;Western medicine and higher dosage traditional Chinese medicine group's rat's kidney medulla swelling.Ultramicrostructure demonstrate middle dosage group of traditional Chinese medicine's renal cells approximately normal;in other preventive groups renal cells there has multus cytolysosome and dimin bioblast,it's functional compensation.3. The therapeutical effect effect of prescription HERBA SIEGESBECKIAE capsule on Hyperuricemia animal modelBased on experiment one,using adenine and Oxonic Acid make the hyperuricemia animal model.Regroup these rat according to it's serum uric acid after three week,using different dosage traditional Chinese medicine and HPPIsopurind intragastric administration,observe blood biochemical indicator and nephridial tissue's structur change on every group rat.1. Middle,high dosage traditional Chinese medicine and HPPIsopurind can decrease serum uric acid, HPPIsopurind is faster,they all have difference by comparing with control group2. Every group of traditional Chinese medicine Can decrease blood-serum creatinine,middle dosage group is faster,have difference by comparing with control group((P<0.05);every therapeutic group's creatinine clearance rate is higher,have significant deviation comparing with control group(P<0.01);lower and middle dosage traditional Chinese medicine group serum urea nitrogen content step down,high dosage traditional Chinese medicine and HPPIsopurind group urea nitrogen step down less, have significant deviation comparing with control group(P<0.01).3. Middle dosage traditional Chinese medicine group renal tubule and acinus renis have perfacte structure,control group, HPPIsopurind and middle dosage traditional Chinese medicine renal cells's structure approximately normal.other groups's cellular structure confused, pathologic state.Conclusion:1. Oxonic Acid and adenine association do well in inducinghyperuricemia animal model on rat.2. Prescription HERBA SIEGESBECKIAE capsule have protective function on hyperuricemia animal model, it can prevent hyperuricemia and the damage to kidney effectively.3. Prescription HERBA SIEGESBECKIAE capsule can decrease serum uric acid effectively in modelanimals, and can repair the uratic nephropathy.
