Background and objectCancer is the significant disease which threaten the human healthy , is the second big cause of human being death ,only next to the heart cranial vascular disease.The research indicates that unbalance of cell's proliferation and apoptosis is one of the important cause developing tumours.The apoptosis is regulated and controlled by the series gene and the expression outcomes,and the gene is divided into two kindsrpromotion gend and inhibition gene.There are also certain connect between the two genes.Among them, Bcl-2 and Bax gene is studied more broaden,deepen and thorough. Bcl-2 is able to inhibit apoptosis, the Bax effect is contrary,the abnomal of Bcl-2/Bax proportion anomaly is that apoptosis and proliferate proportion lose balance,the cell breeds unlimitedly, the balance in the body chaos,leading to tumour occurrence.The selectivity of inducing apoptosis may inhibit the happening of tumour and improve the prognosis of tumour,the theory of apoptosis is guide the radioactive seeds 125I combined with chemical seeds Sinofuan treat tumour.The radioactive seeds 125I combined with chemical seeds Sinofuan treat tumour has unique advantage,the sensitization of two means united kill the tumour cell at high limition.Radioactive seeds 125I combined with chemical seeds Sinofuan inhibit the growth of mice H22 hepatoma cells was observed,the relationship between inhibitory action and apoptosis was discussed,checking the expressions and their interrelation of apoptosis related gene,the theory of radioactive seeds 125I combined with chemical seeds Sinofuan treat tumour was implied.Materials and methods1 Meterialsmice H22 hepatoma cells,radioactive seeds 125I,chemical seeds Sinofuan.2 MethodsThe building-up mice tumour models were implanted radioactive seeds 125I, chemical seeds Sinofuan and radioactive seeds 125I combined with chemical seeds Sinofuan respectively, the speed of growth of tumour was observed, ratio of inhibiting tumour was caculated, compare each group tumour growth condition, adopt HE dyeing and immuocytochemical staining technology,check the expressions of Bcl-2 and Bax in each group.3 Statistical analysisThe experiment data use mean±standard(x±s),statistical method adopt one-way analysis of variance of the SPSS13.0 software,statistically significant level was considered as "alpha equals 0.05".Results1 The curve of tumor growth and ratio of inhibiting tumor in each group of mice with tumorThe tumor growth of I group mice with tumor is quicker,IV group is slowest, weight of tumour is 1.04±0.28g,has significant difference compare to the other three groups (P<0.05).Weight of tumour in II group,III group wasl.69±0.32g,1.71±0.11g respectively,have significant difference compare to contrast group(P<0.05),and there has no significant difference in two groups(P>0.05).2 HE dyeing result in each groupThe nuclear of tumor in I group shrinks,and more cell vacuole degeneration, necrosis,resolving, shrink in other three groups,especially the IV group.3 The immuocytochemical dyeing result of Bcl-2 and Bax in each groupThe positive expressions of Bcl-2 and Bax were mainly localized in the cytoplasm.The color of the positive signal mostly stain yellow,palm fibre,minority stain tan.The positve cell and nuclear is big,the core of nuclear is easy to see and cytoplasm is rich in the cell.The average optical density of Bcl-2 in II group ,III group,IV group is 123.62±2.67,123.67±3.70,93.85±4.68 respectively,the average optical density of Bax is 107.17±1.68,108.68±2.13,136.50±3.94,the ratio of Bcl-2/Bax is 1.15±0.19, 1.24±0.25,0.69±0.03.Compare with I group: average optical density of Bcl-2 in II group,III group,IV group decreased,average optical density of Bax raised,ratio of Bcl-2/Bax also raised,have statistics significance (P<0.05),But there was no statistics significance between II group and III group (P>0.05);compare to II group and III group:the expression of Bcl-2 and Bax,Bcl-2/Bax has statistics significance (P<0.05).Conclusions1 Radioactive seeds 125I combined with chemical seeds Sinofuan has more stronger inhibition totumor cells.2 Chemical seeds Sinofuan sensitizes the radioactive seeds 125I inducing apoptosis in tumor cells.3 The expression of Bcl-2 and ratio of Bcl-2/Bax decreased,the expression of Bax raised is one of mechnism of radioactive seeds 125I combined with chemical seeds Sinofuan inducing apoptosis of tumor cells.
