Background:Health longevity is eternal theme and ultimate challenge in bioscience. Senescence is increasingly critical nowadays, so it is significant to delay aging, prolong healthy time, shorten disease stage and improve the life quality of aged people. Several studies suggest that the centenarians are mainly associated with keeping away from the common age-associated diseases, which is easily affected the other senior citizen, such as cardiovascular disease, diabetes, senile dementia and so on , as well as the maintenance of good immunity condition. Several studies revealed that these age-associated diseases were markedly associated with plasma levels of interleukin-6 (IL-6), tumor necrosis factor alpha(TNF-α) and interleukin-10 (IL-10). Thus these levels could reflect the condition of systemic immunity . Therefore, we hypothesize that the plasma levels of IL-6, TNF-αand IL-10 in the longevity are different from the other aged people and these cytokine levels could reflect the immune status in longevous people.Objectives:To study the association of the plasma cytokine levels of IL-6, TNF-αand IL-10 with the longevous people of Yuxi and Zhanyi in Yunnan province and provide some data and foundation for the geronotologists.Methods:We recruited 117 Han individuals over 90 years in Yuxi and Zhanyi in Yunnan province, 133 longevity' lineal relation as lineal control and 96 individuals as irrelative control, who are the spouses of the lineal control, live in the same area and do not have blood-relationship with the longevous people. The levels of IL-6, TNF-αand IL-10 of blood plasma were measured with double-antibody sandwich ELISA. All statistics were conducted by software SPSS 13. 0.Results:Significantly increased IL-6 levels were detected in the plasma of longevous group when compared with the lineal control and irrelative control (P<0. 017). IL-6 levels were no different in the plasma of the lineal control when compared with irrelative control(P > 0.017). The plasma levels IL-6 of male longevous group were significantly higher than that of female longevous group (P<0. 05), the same was found between female and male irrelative control. TNF-αlevels were no different in the plasma of the longevous group when compared with the lineal and irrelative controls(P > 0.017), TNF-αlevels were no different in the plasma of the male longevous group when compared with female longevous group (P> 0. 05) , neither was found between male and female controls (P > 0.05). IL-10 levels were no different in the plasma of the longevous group when compared with the lineal control (P > 0.017). The plasma levels of IL-10 in longevous group were significantly higher than that of the irrelative control(P <0.017). IL-10 levels were no different in the male longevous group from the female longevous group(P > 0.05), neither was found between male controls and female controls.Conclusions: Significantly increased levels of IL—6 and IL—10 were detected in the plasma of longevous group when compared with the controls. TNF-αlevels were no different in the plasma of the longevous group when compared with the controls, All this partly indicated that the longevity have a low-grade proinflammatory , and further reflected conditions of the longevous' immunity has reached a steady level.
