Objective To explore whether the polymorphism of TryptophanHydroxylase 2 gene (G1463A rs1386494) is associated with schizophrenia in YunnanHan and Jinuo population. Methods PCR-RFLP and ARMS-PCR was used to detectthe polymorphism of Tryptophan Hydroxylase 2 gene (G1463A rs1386494) in 91patients (60 Han and 31 Jinuo) and 93 controls (61 Han and 32 Jinuo), tegother with20 sibling controls of Jinuo. All patients were evaluated with PANSS. Following alinkage disequilibrium test in the 2 loci, association analysis was carried out betweenthe polymorphism, including haplotype, and schizophrenia in all samples. Results Nosignificant difference genotype and allele frequency of the 2 loci was observed inJinuo samples. The genotype and allele frequency of G1463A, also the allelefrequency of rs1386494 was significantly higher in Han patients (P<0.01, P<0.01,P=0.004). But there was no difference in rs1386494 allele frequency in Han samples.The polymorphism of both loci has observed no difference in case-sibling controlstudy of Jinuo. A-G haplotype was observed with a significant higher frequency inboth samples (P=0.033, P<0.000). The distribution of A-A and G-G haplotypes wassignificantly different only in Han samples. 1463A allele carriers among Han patientswere accompanied by a higher score of depression symptoms (P<0.05). Lower scoresin most symptoms were obtained in those carried rs1386494 A allele among Hanpatients (P<0.05). Conclusions There is no association between polymorphism ofTryptophan Hydroxylase 2 gene (G1463A rs1386494) and schizophrenia in Jinuo.rs1386494 genotype and A-A haplotype may possibly associate with schizophrenia inYunnan Han population. 1463A mutation may relate with depression symptom in schizophrenia patients of Yunnan Han. A-G haplotype may play a role inschizophrenia in both population, while, G-G haplotype may possibly participate incertain protection effect in Yunnan Han population.
