The cytotoxicity of different parts of Chinese medicine Longkui (Solanum nigrum L.) were tested, as well as Longkui of different harvesting time, through MTS method. As a result, the whole herb and the fruit exhibited the better cytotoxic activity. Moreover, the cytotoxic activity changes regularly with the different growth periods.The 14 compounds were obtained from fruits and the whole herbs (30% ethanol extracted fraction of macro-porous adsorption resin column chromatography) through various chromatographic techniques. Their structures were elucidated on basis of physico-chemical characters, spectroscopic analysis and chemical communication. The 14 compounds are oleic acid (1), linoleic acid (2), palmitic acid (3),β2-solamargine (4), solamargine (5), degalactotigon in (6), uttroside B (7), syringaresinol-4-O-β-D-glucopyranoside (8), pinoresi -nol-4-O-β-D-glucopyranoside (9), 6-methoxy-7-hydroxycoumarin (10), 3,4-dihydroxybenzo-ic acid (11), p-hydroxybenzoic acid(12), 3-methoxy-4-hydroxybenzoic acid (13), adenosine (14). 8, 9 and 14 were isolated from this genus for the first time. 1, 2, 5, and 6 exhibited the better in cytotoxic activity.The fingerprints of 41 batches of Longkui collected from 33 different regions were studied by HPLC-ELSD method. The better preparation methods of samples were selected through orthogonal experiment. And the chromatography conditions were optimized so that the better resolution and plentiful information were gotten. The similarity of the 41 batches of data was evaluated by the professional analytical software recommended by State Food and Drug Administration (SFDA). On the basis of the common mode of fingerprints, the reference chromatogram was established, the 12 main peaks of which were identified of the reference chromatogram. Moreover, a HPLC-ELSD method for determination of the content of bioactive compounds in the whole herbs was established. The contents of 5 active compounds were determined by this. method. In the end, the bioactivity, the similarity of fingerprints, and the contents were confined to evaluate the quality of the whole herbs in the different harvesting times.
