Objective: To investigate the expression of farnesyltransferase(FTase) beta-subunit mRNA inendometrial cancer tissues and its relationship with point mutation in codon 12 of K-ras.Methods: 43 endometrial cancer specimens and normal endometria were obtained. FTasebeta-subunit mRNA expression was investigated by reverse transcriptase-polymerase chainreaction(RT-PCR). Point mutation in codon 12 of K-ras was detected by polymerase chainreaction-restriction fragment length polymorphism(PCR-RFLP). X~2-Test and spearman was usedto analyse the data of the experiment.Results: The mean levels, of FTase beta-subunit mRNA were significantly higher in theendometrial cancer tissues than those in the matched normal tissues in 43 cases (0.894±0.48 vs0.69±0.40, P=0.014). There was 13 cases of mutation of K-ras 12 codon in 43 cases of cancers (13/43,30.23%), but no mutation in the normal endometria tissues. There was 11 case which showed bothFTase beta-subunit overexpression and K-ras mutation in 43 cases endometrial cancer. K-ras mutationwas founded in 11 of 18 case FTase beta-subunit mRNA overexpression. So significant relationshipwas found between the expression of FTase beta-subunit mRNA and K-ras mutation.Conclusions:①The expression of FTase beta-subunit mRNA is higher in the endometrialcancer tissues than in the normal endometria tissues.②The ratio of point mutation in codon 12 ofK-ras is 30.23%in endometrial cancer.③There is significant relationship between the expression ofFTase beta-subunit mRNA and point mutation incodon 12 of K-ras.
