In this paper, we study electrochemical property of four rhubarb anthraquinones compounds and the interactions between four rhubarb anthraquinones compounds and bovine serum albumin (BSA) by using Electrochemical/Spectral method, suggesting mechanism of the interaction. Electrochemical probe determining protein is filtrated. The paper contains the important content as followed:1. Electrochemical property of four rhubarb anthraquinones compounds (chrysophanol ,Rhein,Emodin and rheochrysidin ) is studied by cyclic voltammetry (CV). Result shows: the other three compounds beside rheochrysidin have a better voltammetric behavior in acid solution, but voltammetric behavior of rheochrysidin is not stability at the same condition. Ep of four rhubarb anthraquinones compounds has a positive relationship with pH in the range of pH 2.8-8.0. Redox reactions are a two electrons two protons process. The compounds are adsorbed on the surface of electrode.2. Mechanism of interaction between four rhubarb anthraquinones compounds and BSA is studied by CV and diferential pulse voltammetry(DPV).The number of electron and proton shifted and the standard electron–transfer rate constant (Ks) of four rhubarb anthraquinones compounds don't change distinctly with and without BSA, and peak current decreases, suggesting that the electrochemical inactive supermolecule is formed. Four rhubarb anthraquinones compounds may act as electrochemical probe of proteins.3. The interaction between rhubarb anthraquinones compounds and BSA is studied by Ultraviolet and visible spectropotometry (UV- vis). The interaction between rhubarb anthraquinones compound and BSA is calculated by Matalab 6.5 lingual authorized program (principle of orthogonal projection). The result shows that all the remains spectrums are nonzero and all the vector module of residue spectrum of the curve of remains are positive number. It proves that a new compound have formed in the reaction system. Finally, coalescent constant of interaction between four rhubarb anthraquinones compounds and BSA is calculated by Scatchard model-equation.
