Serum albumin is the basic composition of all kinds of life activities and the most vitalcarrier protein. The study of interaction between Sal A, Sal B, Rosmarinic acid, Danshensuand BSA is meaningful to understand the transportation, metabolism, medicinal mechanism ofthese drugs in animal’s bodies. Through the fluorescence and UV-visible spectrophotometry, itshows the details of the interaction between bovine serum albumin (BSA) and the four drugswhich are abstracted from salvia on the molecular level. The change of fluorescence intensityof BSA revealed the interaction between drug and BSA. It would be a great guiding for theseimportant water-soluble compounds in Salvia to be designed and synthesized as new drugs.The basic information of salvianolic acids from Radix Salviae miltiorrhizae and BSAwas introduced firstly. Then, the contents, methods and the developments of the research onthe interaction between small molecular of drugs and serum albumin in lately years have beenexpatiated. This thesis was developed based on the apply actuality of fluorescencespectroscopy especially in this research filed.The interaction between Sal A and BSA was studied by fluorescence spectroscopy. It wasrevealed that Sal A was significantly quenched fluorescence of BSA. The binging constantsand thermodynamics parameters of the interaction between Sal A and BSA was calculated. Itshows that hydrogen bond and van der waals force was the main power between Sal A andBSA.The interaction between Sal B and BSA was studied by fluorescence spectroscopy. It wasproved that the fluorescence quenching of BSA by Sal B belongs to dynamic quenching. Thecharacter of the interaction between Sal B and BSA such as fluorescence quenchingmechanism, binding constant, binding site and force type was also investigated at the sametime.Through fluorescence spectroscopy, the interaction between Rosmarinic acid and BSAwas investigated. The way of Rosmarinic acid caused quenching of BSA was discussed. Thebinding constants and thermodynamics parameters was calculated and revealed the type ofmain power between Sal A and BSA. The interaction between Danshensu Salvianic acid A and BSA was studied by UV-Visspectroscopy and synchronous fluorescence spectroscopy at three different temperatures.UV-Vis spectroscopy indicated that Danshensu Salvianic acid A-BSA system generated newcomplex. The fluorescence spectroscopy of Danshensu Salvianic acid A-BSA system showedthat Danshensu Salvianic acid A enhanced the fluorescence intensity of BSA. The data ofsynchronous fluorescence spectroscopy was used to calculated binding constant and the thethermodynamics parameters of the interaction between small molecular of drug and serumalbumin for the first time. The main force between Danshensu Salvianic acid A and BSA wasobtained.
