| Objective: To analysis the expression and significance of transforming growth factorβ3(TGF-β3),matrix metalloproteinase inhibitor 1(TIMP1),B cell lymphoma /leukemia gene 2(Bcl-2)in the nucleus pulposus tissue of degenerative lumbar intervertebral disc. Methods: 140 patients undergoing surgical treatment for L2-L5 lumbar disc herniation department of Orthopedic Surgery of the Via Christi Health Hospital of University of Kansas School of Medicine were selected as the study group, which divided into the mild degeneration group, the moderate degeneration group and the severe degeneration group according to the Videman grading standard, each group with 45、47、48 patients and 17 patients undergoing surgical treatment for L2-L5 lumbar fracture were selected as control group at the same period. The expression of TGF-β3,TIMP1 and Bcl-2 in nucleus pulposus were detected by immunohistochemistry, and the correlation between the expression levels of TGF-β3,TIMP1 and Bcl-2 and the score of Videman were analyzed, the data were processed by statistical software SPSS22.0.Results: Compared with the control group, the Videman scores of the study group was higher, and the mild degeneration group < moderate degeneration group <severe degeneration group(P < 0.05),the mean light density and positive rate of TGF-β3,TIMP1 and Bcl-2 expression in the nucleus pulposus of the study group were lower, and the mild degeneration group > moderate degeneration group >severe degeneration group(P <0.05).The light density of TGF-β3 in degenerative nucleus pulposus was negatively correlated with Videman score(r=-0.929,P<0.05),the light density of TIMP1 was negatively correlated with Videman score(r=-0.961,P<0.05),the light density of Bcl-2was negatively correlated with Videman score(r=-0.947,P<0.05).Conclusions:TGF-β3,TIMP1,and Bcl-2 are expressed in normal and degenerative lumbar intervertebral disc nucleus, and the levels of expression are negatively correlated with the degree of degeneration, may help delay the degeneration of the lumbar intervertebral disc. |
